不同剂量辛伐他汀对慢性心房重构犬心房肌细胞内游离钙离子浓度的影响   [摘要] 目的 探讨不同剂量辛伐他汀剂量对慢性心房心动过速所致心房重构犬心房肌细胞内游离钙离子(Ca2+)浓度的影响。 方法 将28只健康杂种犬随机分为对照组、起搏组、干预1组及干预2组,每组各7只。起搏组、干预组利用快速心房起搏(ATP)建立慢性心房重构模型,干预组ATP前5 d始分别给予口服辛伐他汀10 mg/d(干预1组)和60 mg/d (干预2组)。用激光共聚焦显微镜技术,以Flou-3/AM作为钙指示剂,测定四组犬心房肌细胞内游离Ca2+浓度。 结果 起搏组犬心房肌细胞内游离Ca2+浓度(162)较干预1组(140)、对照组(109)及干预2组(118)均有显著增高,差异有统计学意义(P 0.05)。 结论 大剂量辛伐他汀能更有效抑制慢性心房重构犬心房肌细胞内钙超载。   [关键词] 辛伐他汀;心房重构;钙超载;房颤   [中图分类号] R972 [文献标识码] A [文章编号] 1673-7210(2014)01(c)-0036-03   Effect of the different dose of Simvastatin on concentration of intracellular free Ca2+ in atrial muscle cell of dogs with chronic atrial remodeling   WANG Jian1 PANG Juntao2   1.Department of Cardiology Internal medicine, the Peoples Hospital of Weifang City, Shandong Province, Weifang 261041, China; 2.Intensive Care Unit, the Peoples Hospital of Weifang City, Shandong Province, Weifang 261041, China   [Abstract] Objective To investigate effect of the different dose of Simvastatin on concentrations of intracellular free Ca2+ in atrial muscle cell of dogs with atrial remodeling caused by chronic atrial tachycardia. Methods 28 healthy mongrel dogs were randomly divided into control group, pacing group, therapeutic group 1, and therapeutic group 2, with 7 dogs in each group. Models of chronic atrial remodeling were set up induced by rapid atrial pacing (ATP) in pacing group and therapeutic group. 10 mg Simvastatin was administered orally 5 days before ATP in therapeutic group 1, 60 mg in therapeutic group 2. Intracellular free Ca2+ concentrations were measured in dogs atrial muscle of four groups with laser confocal microscopy and with Flou-3/AM as calcium indicator. Results Concentration of intracellular free Ca2+ in dog atrial muscle cell of pacing group (162) increased significantly than that of therapeutic group 1 (140), control group (109) and therapeutic group 2 (118), the differences were statistically significant (P 0.05).
