ThetransplantabletumorsofLV-APE1-shRNAgroupgrewslowly，thevolume（0.467±0.052）cm3andweight（0.267±0.082）gwerelowercmparedwithLV-APE1-NC group （1.700±0.063）cm3、（1.018±0.160）g and control group（1.780±0.095）cm3、（1.107±0.178）g(P0.01).Thetumorgrowthinhibitoryratewas 73.772%.necrosiswasobservedinfrequentlyinLV-APE1-shRNAgroupbyHEdyeunderlightmicroscope，butobservedfrequentlyinLV-APE1-NCgroupandcontrolgroup. Apoptosissmallbodywasmorecomparedwiththeothertwogroups.Pathologicfissionphase＜fiveperhighpowerfieldinLV-APE1-shRNAgroupandPathologicfissionphase＞fiveperhighpowerfieldinLV-APE1-NCgroupandcontrolgroup.ItwasshowerthatthemRNAexpressionlevelsofAPE1，c-Fos，c-Jun，caspase-3andMMP-2werelowercmparedwithLV-APE1-NCgroupandcontrolgroup(P0.01).AfterAPE1genesilence，mRNAexpressionlevelsofAPE1decreasedby90.622%，c-Fosdecreasedby47.580%，c-Jundecreasedby47.402%，caspase-3decreasedby65.355%，MMP-2decreasedby64.369%。Andtheirexpressionswerepositivecorrelation.Itwas showed that the protein expression levels of APE1，c-Fos，c-Jun，caspase-3andMMP-2werelowercmparedwithLV-APE1-NCgroupandcontrolgroup(P0.01).AfterAPE1genesilence，proteinexpressionlevelsofAPE1 decreasedby72.439%，c-Fosdecreasedby71.872%，c-Jundecreasedby80.399%，caspase-3 decreased by 75.616%，MMP-2 decreased by 88.393%。And theirexpressionswerepositivecorrelation.ItwasfoundthatapoptosiscountofLV-APE1-shRNAgroupwashighercomparedwiththeothertwogroups（P0.01），theapoptosisindexof LV-APE1-shRNAgroup was5.503％，LV-APE1-NCgroupwas2.911％andcontrolgroupwas2.569％byTUNELmethod.ConclusionsItwassuccessfultoestablishanxenogrftmodeloftheHumanHepatocellularCarcinomaCelllineMHCC97-H.APE1genesilenceofMHCC97-Hbyinfectedlentivirusvectorcaninhibitobservablythegrowthoftransplanttumor.RT-PCR assay and Western blot assay showed that the mRNA and proteinexpressionlevelsofAPE1，c-Fos，c-Jun，caspase-3andMMP-2wereinhibitedobservably，andAPE1canregulatexpressionofc-Fos，c-Jun，caspase-3andMMP-2 positively.APE1couldhaveahandinproliferation，invasionandmetastasisofhumanhepatocellularc
