And analyze the relationship between methylation of promoter and mRNA expression of Annexin Ⅱ.Methods All specimens of 45 of LSCC, 18 of nontumorous lung tissue (NLT) （,serveas normal control）were pathological confirmed and the expression level weredetected by immunohistochemistry S-P method，and the correlation between the expression of Annexin Ⅱ and clinic factors of LSCC were analysized by SPSSsoftware.The DNA and RNA were extracted from 6 cases of biopsy specimens of LSCC and 2 cases of biopsy specimens of NLT, respectively. And the methylation ofAnnexin Ⅱ were detected by Methylmion Specific PCR (PCR-MSP) and mRNAlevel were detected by RT-PCR. And analysis the correlation between methylation of Annexin Ⅱ and the expression of mRNA of Annexin Ⅱ in LSCC .Results 1) The positive ratio of expression of Annexin Ⅱ were 68.9%(31/45) in LSCC, 94.4%(17/18) in NLT. The expression level of NLT were higher than that of LSCC. And there were significantly difference between LSCC and NLT (P＜0.05).The positive ratio of Annexin Ⅱexpression were 90%（9/10）、80.9%(17/21)、35.7%（5/14） in well-differentiated, middle-differentiated and poor-differentiated LSCC, respectively.
