AbstractNanoliposomesasacarriercausedwideattentionandresearchforitsuniquefeature.Notonlyitsphospholipidssusceptibletobeoxidized,easyintegrationgathered,theseunstablefactorsmakingiteasiertocausethedrugleakage,butalsolackofinitiativetargeting,soitsapplicationisrestricted.CurcuminExtractedfromcurcuma,hasaspectrumofbiologicalactivities.However,becauseofthepoorsolubilityinwater,sensitivetolight,heatandotheradverseenvironmental,itsbioavailabilityisverylow.Usingliposomewrappeditup,thebioavailabilityofcurcumincanbeextentlyimproved.Inordertofurtherimprovethephysicalstability,sustainedreleaseandcellularuptakerateofliposomes,themaincontentofthisstudyincludemakethesmallmoleculesoffolicacid(fa)conjugatedwithchitosan(cs),Folicacid-chitosan(fa-cs)coatedonthesurfaceofcurcuminliposomes(lip-cur)byelectrostaticinteractionforce.Sothefolicacid-chitosancoatedliposomes(fa-lip-cur)wasformed.Theninvestigatethedifferencesofstoragestability,slow-releasingpotentialandcellularuptakeproportybetweenfa-lip-curandlip-cur.BasedonEDCandNHSascatalyst,folicacid-chitosanwassynthesized.Afterinfraredspectrummethodisusedtodetectthefunctionalgroupsofreactionproduct.Accordingtotheabsorptionpeakof1656cm–1and1606cm-1,theyrepresentthecharacteristicpeakofchitosanandfolicacidrespectively.Theresultsshowthatfolicacid-chitosanwasformedsuccessfully.Bythinfilmdispersionmethodcombinedwithdynamichighpressuremicrojetpreparednanoliposomesofcurcumin(lip-cur).Thesizeofliposomeswas67.4±3.4nm,zetapotentialwas-13.81±2.75mv,andtheparticlesizeandzetapotentialoffa-lip-curwere103.6±4.1nmand16.35±3.54mvrespectively.TheresultofSlowreleaseinvitroexperimentshowthatfa-lip-curshowbetterslowreleaseeffectunderalkalescentconditionwhichcomparedwithlip-cur.After24h,thedrugonlyrelease17.7%fromfa-lip-cur.Twokindofliposomesstoragefor4weeksundertheconditionof25℃,theirparticlesizeandzetapotentialchangeweekly,butfa-lip-curshowsbetterprotectivedrugsthanlip-cur.Atthesametime,weinvestigatetheaffinitybetweenfa-lip-cur/lip-curandCaco-2.DeterminedbyMTTc
