IL-24 联合顺铂对裸鼠宫颈癌淋巴道转移模型的影响摘要目的：建立裸鼠人宫颈癌 Siha 细胞淋巴道转移模型，探索 IL-24 基因联合低剂量的顺铂 对裸鼠宫颈癌淋巴道转移模型的淋巴转移的影响，并证实其通过 HMGB1/ RAGE 信 号转导通路完成对宫颈癌细胞淋巴转移的调节和抑制作用。方法：将人宫颈癌 Siha 细胞分别接种于 42 只裸鼠一侧后足的爪垫，成功建立裸鼠人宫 颈癌 Siha 细胞的淋巴道转移模型后，随机分为 6 组，每组 7 只。分别用 PBS 缓冲液、 pDC316 空质粒、pDC316-h IL-24 基因重组质粒、pDC316-hIL-24 基因重组质粒联合 低剂量顺铂、低剂量顺铂、高剂量顺铂进行干预治疗，观察裸鼠的一般状态的变化及 淋巴转移情况，采用病理学方法认证淋巴结及淋巴结内部的宫颈癌浸润，应用实时定 量 PCR 检测裸鼠的组织中 IL-24 基因的表达情况，采用蛋白印迹法及免疫荧光法检 测裸鼠的淋巴结中 HMGB1/ RAGE 信号通路的表达情况。结果：1.各组裸鼠人宫颈癌 Siha 细胞淋巴道转移模型干预治疗前的差别无统计学意义；2.各组裸鼠人宫颈癌 Siha 细胞淋巴道转移模型干预治疗后的淋巴转移个数差别有统 计学意义,IL- 24 基因联合低剂量顺铂组对裸鼠模型的淋巴转移的抑制率明显高于其 他各组；3.病理学方法证实取材部位为淋巴结，并有癌细胞浸润；4.实时定量 PCR 检 测 IL-24 基因成功转染进入裸鼠体内；5.干预治疗后各组裸小鼠的 HMGB1/ RAGE 信 号通路的荧光表达结果的有效率有差别，联合组的荧光表达明显少于其他各组；6. 干预治疗后蛋白印记方法检测各组裸小鼠的淋巴中 HMGB1/ RAGE 信号通路的蛋白 表达差别有统计学意义，联合组的蛋白表达明显少于其他各组。结论：pDC316IL-24 联合顺铂治疗可明显抑制宫颈癌的淋巴转移，通过 HMGB1/ RAGE 信号通路完成对宫颈癌细胞淋巴转移的抑制作用，证实 HMGB1/ RAGE 信号通路参 与宫颈癌细胞淋巴转移的作用机制，并实现对淋巴转移的抑制作用。关键词：白细胞介素类；顺铂；HMGB1；宫颈癌；淋巴转移Cisplatin of IL-24 affect the lymphatic metastasis on the cervical cancer nude modelAbstractObjective:To explore the influence of IL-24 in combination with cisplatin on the cervical cancer nude model of lymphatic metastasis, confirmed the completion of lymph node metastasis in cervical cancer cells through the regulation of HMGB1 / RAGE signaling pathways.Methods:Siha cells were seeded in 42 side of the rear paw-pads of nude, the nude model of lymphatic metastasis was successfully established and randomly divided into six groups , each 7.
