摘要IGF-Ⅰ、OPN、PTEN在非小细胞肺癌中的表达及其意义研究生：周颖导师：买买提艾力.吾马尔副教授摘要目的探讨胰岛素样生长因子Ⅰ（IGF-Ⅰ）、骨桥蛋白（OPN）和PTEN在人非小细胞肺癌中的表达及其相关性，并分析其与非小细胞肺癌发展预后的关系。方法应用免疫组织化学SP法检测61例肺癌组织和30例对照正常肺组织中IGF-Ⅰ、OPN、PTEN蛋白的表达情况。同时应用原位杂交方法检测61例肺癌组织和30例对照正常肺组织中IGF-ⅠmRNA、OPNmRNA的表达情况。结果61例肺癌组织中IGF-Ⅰ、OPN蛋白、IGF-ⅠmRNA、OPNmRNA表达的阳性率显著高于正常肺组织（P＜0.05）；IGF-Ⅰ蛋白以及IGF-ⅠmRNA的表达与肿瘤分化程度、淋巴结转移密切相关（P＜0.05）；OPN蛋白以及OPNmRNA的表达与肿瘤分化程度、淋巴结转移、TNM分期密切相关（P＜0.05）；61例肿瘤组织中PTEN蛋白表达的阳性率明显低于正常肺组织（P＜0.05），PTEN蛋白表达与肿瘤大小、肿瘤分化程度、淋巴结转移有关（P＜0.05）。IGF-Ⅰ蛋白的表达与OPN蛋白的表达呈正相关（P＜0.05），IGF-ⅠmRNA的表达与OPNmRNA的表达呈正相关（P＜0.05），IGF-Ⅰ、OPN与PTEN的表达呈负相关（P＜0.05）。结论IGF-Ⅰ、OPN、PTEN与非小细胞肺癌的发生发展密切相关，三者的表达对判断肺癌恶性程度及预后具有参考意义。关键词：非小细胞肺癌；胰岛素样生长因子Ⅰ；骨桥蛋白；PTEN；免疫组织化学；原位杂交新疆医科大学医学硕士学位论文SignificanceofExpressionofIGF-Ⅰ、OPNandPTENinNon-smallcellLungCancerPostgraduate:zhouyingSupervisor:Prof.maimaitiaili.wumaerAbstractObjectiveToinvestigatetheexpressionofIGF-Ⅰ、OPNandPTENanditscorrelationwithmetastasisandprognosisinNon-smallcellLungCancer.MethodsImmunohistochemicalstainingwasusedtodetecttheexpressionofIGF-Ⅰ、OPNandPTENin61casesofNon-smallcellLungCancerand30casesofadjacentnormaltissuesfrompatientswithNSCLC.InsituhybridizationwasusedtodetecttheexpressionofIGF-ⅠmRNAandOPNmRNAin61casesofNon-smallcellLungCancerand30casesofadjacentnormaltissuesfrompatientswithNSCLC.ResultsIn61casesofNon-smallcellLungCancer,theexpressionofIGF-Ⅰ、OPN、IGF-ⅠmRNAandOPNmRNAwerehigherthanthoseintheadjacentnormaltissues(P＜0.05).ThepositiveratesofIGF-ⅠandIGF-ⅠmRNAwerecloselycorrelatedwiththecellulardifferentiationandlymphnodemetastasis(P＜0.05).ThepositiveratesofOPNandOPNmRNAwerecloselycorrelatedwithcellulardifferentiation,lymphnodemetastasisandTNMstaging(P＜0.05).In61casesofNon-smallcellLungCancer,theexpressionofPTENwaslowerthanthoseintheadjacentnormaltissues(P＜0.05),ThepositiveratesofPTENwascoselycorrelatedwithsizeoftumor,cellulardifferentiationandlymphnodemetastasis(P＜0.05);TheexpressionofIGF-ⅠwascorrelatedpositivewiththatofOPN(P＜0.05);TheexpressionofIGF-ⅠmRNAwascorrelatedpositivewiththatofOPNmRNA(P＜0.05);Theexpressi
