conclusionathomeandabroadisnotconsistent.Objective：OurstudyonidentificationofApoEgeneassociatedwithTNF-α-850 genewaspurposedtoinvestigatewhethertherewereApoEandTNF-α-850 genepolymorphismsornotandthefrequenciesofgenotypeandalleleindifferentpopulations.Aswellas,ourobjectiveofthestudywastoanalysetheassociationofapolipoproteinEandtumournecrosisfactor-α(TNF-α)genepolymorphismsatposition-850C/TinChinesepopulationofGuangdongprovincewithAlzheimer’s diseaseandtoexplorethegeneticbackgroundofAD.WeexpectthatourstudywillprovidethefoundationofearlydiagnosisandearlyselectionofAlzheimer’sdisease.Method：Weconductedacase--controlstudy.37casesADpatientsand72casesnormalcontrolswereenrolled.GenomeDNAfromtheperipheralvenous bloodwastakenrespectively.TNF-αgenepolymorphismsatposition-850C/Tweredetermined by polymerase chain reaction-restriction fragment lengthpolymorphism(PCR-PFLP)method.ApplySPSS17.0statisticalanalysissoftwaretoreadallelesandgenotypes of individual samples，and eachgroupallele frequenciesandgenotypecomparedwithchi-squaretestandlogisticregressionanalysis.ThevalueofPlessthan0.05isconsideredstatisticallysignificant.Result：ADgroupandnormalcontrolsweresimilarinageandsex，subgroups werematehedbyage,sex,diseaseduration.The distributions of ApoE and TNF-αgenetypes followed the law ofHardy-Weinbergequilibrium.IneitherADgroupornormalcontrolgroup,thealleleofApoEε3wasmostcommonandgenotypeofε3/3wasmostfrequentlyseeninChinesepolulationofGuangzhouprovince.TheApoEε4frequencywas significantlyhigherinADpatientsthanthatinthenormalcontrols(P＜0.05).Theageandsex-adjusted,OddsratiofortheriskofADincarriersofApoEε4allelewas 5.333（OddsRatio=5.333,95%confidenceinterval:2.054-13,851,P=0.001）. ThedistributionofApoEε2allelefrequencyinADgroupandnormalgroupwasno significantdifference(P＜0.05).OurdatashowedthatthedistributionofeveryTNF-α-850genotypeandallelewerenosignificantlydifferentinADpatientscomparedwithnormalcontrols(P＞0.05).Inaddition,associatedwiththeApoEε4allele,thedistributionof
