曲妥珠单抗 binds to subdomain IV of the HER2 extracellular domain and exerts its antitumor effects by blocking HER2 cleavage, stimulating antibody-dependent, cell-mediated cytotoxicity and inhibiting ligand-independent, HER2- mediated mitogenic signaling. Pertuzumab, a humanized monoclonal antibody that binds HER2 at a different epitope of the HER2 extracellular domain (subdomain II) than that at which 曲妥珠单抗 binds. Pertuzumab prevents HER2 from dimerizing with other ligand-activated HER receptors, most notably HER3. Like 曲妥珠单抗, pertuzumab stimulates antibody-dependent, cell-mediated cytotoxicity. Because of their different bindings sites, 曲妥珠单抗 and pertuzumab have complementary mechanisms of action. Whereas 曲妥珠单抗 blocks HER2 cleavage and inhibits ligand-independent signalling, pertuzumab exerts its effects by inhibiting ligand-dependent signalling, particularly between HER2 and HER3, which is known to activate a potent cell survival and proliferation signal.
