IL-24 基因联合顺铂对人宫颈癌 Siha 细胞淋巴转移抑制作用的 研究摘要目的：研究IL-24基因与顺铂（DDP）联合应用后，对宫颈癌裸鼠移植瘤淋巴转移的抑 制作用。方法：建立宫颈癌Siha细胞裸鼠的淋巴转移模型；据治疗方案将动物模型分为 5 组： A组：PBS 组、B组：pDC316空质粒组、C组：IL-24 组（pDC316-hIL-24 质粒）、 D组：顺铂组、E组：联合组（pDC316-hIL-24 质粒联合顺铂）。观察并记录裸鼠的 一般情况、肿瘤生长及淋巴转移情况，用PCR (聚合酶链反应)法鉴定移植瘤中IL-24 的表达；免疫组化法证实所取组织为转移的淋巴结；用Western blot 法（蛋白质印 迹法）检测各组转移淋巴结中的小窝蛋白（Caveolin-1）及VEGF-D表达水平，通过 比较各组小窝蛋白（Caveolin-1）及VEGF-D的表达水平以及各组之间淋巴结转移数， 评价 IL-24 基因联合顺铂对人宫颈癌Siha细胞淋巴转移的影响。结果：重组质粒 pDC316-hIL-24 成功转染并表达；所取组织为转移的淋巴结；干预 后联合组淋巴转移总数最低（1.57±0.79），差别有统计学意义（P＜0.05）；联合组下 调 VEGF-D（VEGF-D 为0.134±0.005）和上调Caveolin-1（Caveolin-1 为0.790±0.006） 的表达效果最明显。结论：IL-24 基因联合顺铂可通过下调 VEGF-D和上调Caveolin-1 的表达水平进而 抑制 Siha 肿瘤细胞淋巴转移能力，优于单独使用顺铂的抑制淋巴转移效果；2.
