精品论文 参考文献 脐血间充质干细胞移植治疗肝硬化大鼠的疗效及归巢特性观察 湖南省人民医院 湖南长沙 410005 【摘 要】目的：观察人脐血间充质干细胞对肝硬化大鼠的疗效及其归巢特性。方法：采用四氯化碳法制备肝硬化大鼠模型。实验随机分为三组，A组（n=20）：肝硬化大鼠经门静脉注射人脐血间充质干细胞1ml （5times;106个）；B组（n=20）：肝硬化大鼠经门静脉注射等体积的PBS；C组（n=10），正常大鼠经门静脉注射人脐血间充质干细胞1ml。细胞移植后4周，取各大鼠静脉血及肝脏组织进行检测。结果：A组与B组比较，细胞移植后4周后大鼠的血清谷丙转氨酶、谷草转氨酶、总胆红素降低，白蛋白显著升高（Plt;0.01）；透明质酸、层粘连蛋白、前胶原蛋白Ⅲ及胶原蛋白Ⅳ水平降低（Plt;0.01）；肝细胞炎性坏死、脂肪变及肝纤维化有不同程度改善（Plt;0.05或Plt;0.01）。免疫组化染色显示：A、C组大鼠肝组织均有人脐血间充质干细胞的定植，但A组定植细胞数明显多于C组（Plt;0.01）。C组大鼠脾、肺、肾等组织均有少量人脐血间充质干细胞定植。结论：人脐血间充质干细胞移植可部分改善肝硬化大鼠的肝功能及病理损伤，肝损伤情况下人脐血干细胞具有归巢特性。 【关键词】肝硬化；脐血间充质干细胞；肝功能；肝纤维化；归巢；定植 【Abstract】Objective：To observe the curative efficacy of human umbilical cord blood mesenchymal stem cells transplantation on liver cirrhosis rats and observe the homing characteristics. Methods A cirrhotic rat model was established by CCL4。The experiments were randomly divided into three groups：group A（n=20），Portal vein injection of human umbilical cord blood mesenchymal stem cells 1ml （5x106）in cirrhotic rats，group B（n=20）cirrhotic rats was injected with the same volume of PBS through portal vein，group C（n=10）was normal rats，through portal vein was injected human umbilical cord blood mesenchymal stem cells 1ml. After 4 weeks of transplantation，vein blood and liver tissue of each rats were obtained for testing. Results At 4 weeks after cell transplantation，group A compared with the group B，levels of serum alanine aminotransferase，aspartate aminotransferase and total bilirubin were significantly decreased，while the albumin level was increased significantly （Plt; 0.01），hyaluronic acid，laminin，collagen III and collagen IV levels were significantly reduced （Plt;0.01），the liver cell inflammatory necrosis，steatosis and liverfibrosis were varying degrees of improvement （P lt;0.05 or P lt;0.01）.
