鸡卵类粘蛋白的分离纯化、活性及分子量测定 （河北大学生命科学学院2012级 生物工程，河北 保定，071002） 【摘要】： 学习本实验的主要目的是掌握蛋白质的提取、分离、纯化及性质测定技术，掌握离心机、分光光度计、核酸蛋白检测仪、电泳仪等实验仪器的使用。 先通过三氯乙酸-丙酮除去蛋清中的杂蛋白，后经等电点沉淀得到类粘蛋白的粗品;然后经过层析柱的装柱，平衡，上样，洗脱，再平衡等步骤实现对类粘蛋白的纯化;下一步，用BAEE法对类粘蛋白活性进行测定，得到类粘蛋白对胰蛋白酶的抑制活性；最后，通过SDS聚丙烯酰胺凝胶电泳检测类粘蛋白的纯度及分子量。 通过本实验的学习得出类粘蛋白是一种专一性很强的胰蛋白酶抑制剂，胰蛋白酶能水解酯键,酰胺键和肽键,利用这一性质用人工合成的底物或天然的蛋白质可以测定胰蛋白酶的活力,由此可计算类粘蛋白的活力。 【关键词】：鸡卵类粘蛋白 纯化 胰蛋白酶 分子量 抑制剂 中图分类号：Q5-33 文献标志码：A The purification and activity and molecular weight determination of CHOM College of Life Hebei University 2009 Bioengineering, Hebei Baoding ,071002 Abstract: The main purpose of learning this experiment is master the technology of protein extraction, isolation, purification and properties mensuration, Master the usage of centrifuge, spectrophotometer, nucleic acidprotein detector, electrophoresis apparatus and so on. First, remove the miscellaneous protein in egg white by TCA – acetone, then get the rough protein by isoelectric precipitation.Then after chromatography column packing column, balance, Sample , elution, rebalance etc to achieve the purification of chicken ovomucoid. Next, use BAEE method to determine the vitality of chicken ovomucoid, get inhibitory activity from chicken ovomucoid with respect to trypsin. Finally, through SDSpolyacrylamide gel electrophoresis to determine the purity and molecular weight of chicken ovomucoid. Through the experiment, we conclud that chicken ovomucoid is a kind of specificity strong trypsin inhibitors, can hydrolyze ester keys, amides keys and peptide bond. Use this nature and synthetic substrate or natural protein can determine the vitality of trypsin, so can calculating the vitality of chicken ovomucoid. Key words：chicken ovomucoid purification trypsin molecular weight inhibitors 前言 鸡卵类粘蛋白（chicken ovomucoid 简称CHOM）是鸡卵清中含有的一种糖蛋白，在电泳行为上常出现不均一性，这主要与糖蛋白的分子量不均一性有关。鸡卵类粘蛋白具有强烈抑制胰蛋白酶的作用，是胰蛋白酶的天然抑制剂，胰蛋白酶抑制剂是一类具有胰蛋白酶抑制作用的物质，该类物质的来源不同 ,其活性和作用的酶也不同，常用于胰蛋白酶的酶学性质的研究[1]。另外对牛和猪的胰蛋白酶有强烈的抑制作用，对枯草杆菌蛋白酶也有一定的抑制作用，但对胰凝乳蛋白酶无抑
