Abstract Objective: To explore the clinicopathological significance in gastric cancer through examining and comparing expressed IL-6 and constitutively activated Stat3 in cancerous and adjacent non-cancerous tissues, as well as the impact of IL-6/Stat3 pathway on the prognosis of patients with gastric cancer. Methods: (1) We chose 181 cases of gastric cancer and adjacent tissues 181 cases, all of samples were the formalin-fixed and paraffin embedding tissues. (2) Immunohistochemistry (SP method) was used to detect the expression of IL-6 and p-Stat3. (3) We collected 181 cases of clinical data and follow-up all the patients, 118 cases were followed. (3) PCR experiment was used to detect the expression of IL-6 and Stat3; (4) Statistical analysis was performed using SPSS (version 17.0) statistical software package. Qualitative data were analyzed using chi-square test. Spearman rank correlation test was used to analyze the correlation between IL-6 expression and Stat3 activation in different tissues. Survival durations were calculated using the Kaplan-Meier method. Results: (1) The expressions of IL-6 and Stat3 in gastric carcinoma tissues are significantly higher than those in adjacent tissues, and they are gradually increased in normal gastric mucosa, chronic atrophic gastritis, low-grade and high-grade squamous intraepithelial neoplasia lesion. (2) The expressions of IL-6 in gastric carcinoma tissues become significantly higher along with the age to older, the depth of invasion to deeper, lymph node metastasis, clinical stage to higher. The expressions of Stat3 become significantly higher along with the depth of invasion to deeper, lymph node metastasis, distant metastasis and clinical stage to higher.
