 n 雌二醇对乳腺癌组织内成纤维细胞生长因 子分泌的影响及意义# 李治，黄韬** 5 10 15 （华中科技大学附属协和医院，武汉，430022） 摘要：目的：探讨内分泌微环境影响雌激素受体表达不同的乳腺癌细胞增殖的作用机制。方 法：通过流式细胞术从乳腺癌组织中获取成纤维细胞，加入雌二醇后进行原代培养后，测定 癌源性成纤维细胞中基质细胞源性因子-1、肝细胞生长因子、胰岛素样生长因子-1 和表皮生 长因子 mRNA 的表达，以及其培养上清对乳腺癌细胞 MCF-7 和 MDA-MB-231S 体外增殖的 影响。结果：加入雌二醇后，癌源性成纤维细胞中基质细胞源性因子-1、肝细胞生长因子、 胰岛素样生长因子-1 和表皮生长因子 mRNA 的表达分别上升到 4.86±1.59 倍、1.91±1.12 倍、1.74±0.68 倍（P 均＜0.01）、1.07±0.35 倍（P＞0.05）。不含和含雌二醇培养的癌源 性成纤维细胞上清浓缩后，加入乳腺癌细胞中体外培养 5 天后，MCF-7 的增殖分别上升了 2.13±0.24 倍和 3.47±0.36 倍（P 均＜0.01），MDA-MB-231S 的增殖分别上升了 1.97±0.22 倍和 3.16±0.31 倍（P 均＜0.01）。结论：雌二醇能够通过上调癌源性成纤维细胞分泌基质 细胞源性因子-1、肝细胞生长因子、胰岛素样生长因子-1，对雌激素受体表达不同的乳腺癌 细胞起间接的促增殖作用，其中基质细胞源性因子-1 可能有着关键性作用。 关键词：乳腺肿瘤；雌二醇；基质细胞源性因子-1 中图分类号：R737.9 20 The Significance of Estradiol Impact on Growth Factor Secretion From Fibroblast in Breast Cancer Tissue LI Zhi, Huang Tao (Union Hospital,Hua Zhong Science and Technology University,Wuhan,430022) 25 30 35 40 Abstract: Objective: To approach the mechanism of endocrine microenvironment impact on proliferation of breast cancer cell with diverse estrogen receptor expression.Methods:To obtain fibroblasts from breast cancer tissues by flow cytometry, and determine stromal cell-derived factor-1, hepatocyte growth factor, insulin-like growth factor-1 and epidermal growth factor mRNA expression in cancer-derived fibroblasts after the primary culture with adding estradiol, as well as the effect of its culture supernatant impact on proliferation of the breast cancer cell line MCF-7 and MDA-MB-231S in vitro.
