目录摘要························································································1第一部分摘要·······················································································1第二部分摘要·······················································································3Abstract4正文第一部分野生型VHL基因提高肾细胞癌细胞786-O对TRAIL敏感性的基础研究序言································································································7材料与方法····························································································9实验结果·····························································································26讨论································································································33结论和展望··························································································46参考文献·····························································································48第二部分综述···································································································59参考文献·····························································································64附录1攻读博士学位期间发表的论文目录······································70附录2主要英文缩略词表·······························································72致谢······························································································74中文摘要第一部分实验研究野生型VHL基因提高肾细胞癌细胞株786-O对TRAIL敏感性的基础研究目的:91%的散发性肾细胞癌患者中存在肿瘤抑制基因vonHippel-Lindau(VHL)的突变或失活。VHL基因以及翻译的产物pVHL能够在转录和翻译水平调节多种与肾细胞癌发生发展相关基因的表达,但与TRAIL介导的肿瘤细胞凋亡之间的关系并不明确,本次研究的目标是在肾细胞癌786-O细胞株上明确野生型VHL基因在TRAIL介导的肿瘤细胞凋亡中的作用。方法:我们选用肾细胞癌786-O细胞株、单克隆携带野生型VHL基因全长DNA的786-O/VHL细胞以及单克隆携带VHL基因不同类型突变DNA的786-O细胞,采用不同浓度的人重组TRAIL和40ug/ml的TRAIL作用不同的时间,利用MTT和流式细胞术来检测上述细胞株对TRAIL介导肿瘤细胞凋亡的敏感性以及野生型VHL基因是否具有提高786-O细胞对TRAIL敏感性的能力。RT-PCR和Western-Blot用于分析死亡受体DR4、DR5以及凋亡抑制蛋白c-FLIL)在转录和翻译的表达水平。为了准确阐明c-FLIP(L)和TRAIL介导786-O/VHL细胞凋亡的相关性,我们用脂质体2000将c-FLIP(L)转染到对TRAIL敏感的786-O/VHL细胞中,观察高表达的c-FLIP(L)和TRAIL敏感性的关系。同时我们利用DR4和DR5的拮抗抗体来观察两种死亡受体和野生型VHL基因的关系。结果:MTT检测
