缩小（p0.05）。结论：（1）IL-21具有抑制人胃癌细胞SGC-7901的增殖、迁移，并促进其凋亡的作用；（2）IL-21与化疗药物5-Fu常规用量联合使用时，上述作用得到协同加强，联合使用IL-21有利于提高单纯使用5-Fu的抗肿瘤效果；（3）IL-21与化疗药物半量5-Fu联合使用时，抗肿瘤细胞增殖的作用仍然存在，但抑制迁移和促进凋亡的作用稍逊于单纯5-Fu常规化疗，但该方案减少了5-Fu的用量，对于减轻化疗的毒副作用有积极意义，在抗肿瘤治疗中的实际应用价值值得进一步探讨；（4）IL-21与5-Fu联用对SGC-7901细胞的凋亡促进作用，与几种重要的凋亡相关蛋白有关，其中可使caspase-3、caspase-8的表达升高，bc1-2的表达降低，c-myc的表达显著增加。【关键词】IL-21，癌，药物疗法，联合，免疫IIIAntitumoreffectsofIL-21andcombinationwith5-FuonhumangastriccancercellanditsmechanismMajor:clinicallaboratorydiagnosisPostgraduate:wushenSupervisor:jianghongmei[Abstract]Objective:Toevaluatetheeffectsandmechanismofcellproliferation,migrationpotentialityandapoptosisonhumangastriccancercellSGC-7901inducedbyIL-21andcombinationwith5-Fu.Methods:(1)SGC-7901cellswerecultured.SGC-7901cellsweretreatedwithdifferentgroups:thegroupIL-21（IL-21100ng/ml）,thegroup1/25-Fu+IL-21（5-Fu25μg/ml+IL-21100ng/ml）,thegroup5-Fu（5-Fu50μg/ml）,thegroup5-Fu+IL-21（5-Fu50μg/ml+IL-21100ng/ml）,for24h,48h,respectively.TheinhibitoryrateofcellproliferationwasdetectedbyMTTassay.(2)Scratchingtestwastakentoevaluatethemigrationpotentialityofgastriccancercellswithdifferenttreatments.(3)Theexpressionofthecaspase-3,caspase-8,bcl-2andc-mycproteininhumangastriccancercelltreatedwithdifferentgroupsbyWesternBlotassay.Results:(1)Comparedwiththegroup5-Fu,inhibitoryeffectofthegroupIL-21and1/25-Fu+IL-21weremarkedlylowerat24hand48h,butat48h,thereweresignificantdifferencesbetweenthegroup5-Fuand1/25-Fu+IL-21(p0.05).ThecombinationofIL-21and5-Fushowedanmarkedlyhigherinhibitoryeffectthanthegroup5-Fuboth24hand48h(p0.001).(2)ThescratchingtestshowedthatallofthefourgroupsmadeaverysignificantinhibitoryeffectonthemigrationpotentialityofSGC-7901cells.At24hand48h,migrationdistancesofthegroupIL-21and1/25-Fu+IL-21wereshorterthanthegroup5-Fu(p0.001).Themigrationdistancesbetweenthegroup5-Fu+IL-21andthegroup5-Fushowedmarkeddifferences(p0.001).(3)TUNELtestshowedthatthegroupIL-21,1/25-Fu+IL-21,5-Fuand5-Fu+IL-21madeaverysignificantcellapoptosiseffectonSGC-7901cells,andtheeffects
