RelationshipbetweenHLA-DQA1polymorphismsandasthmasandthelevelofIFN-γ，IL-4inbloodserumAbstractObjective:ToinvestigatetheassociationbetweenhumanleukocyteantigenDQA1gene(HLA-DQA1)polymorphismsandchildhoodbronchialasthmaandtoexplorewhatimpactofHLA-DQA1genetypehasonchildhoodasthma.AndtoobservethelevelsofIL-4,IFN-γinperipheralbloodofasthmaticpatientsandanalyzetherelationshipbetweentherelatedHLA-DQA1geneandthelevelsofIL-4,IFN-γ,whichcanbeusedtoprovidetheobjectiveevidenceinstudythepathogenesisofasthmaandmethodsforthepreventionandcontrolofasthma.Methods:1.Serumcollection:(1)asthmagroup:45casesofchildrenasthmainacuteepisodewerestudied.(2)controlgroup:60casesofnormalchildrenwerechosentobecontrolgroup.2.Detectindex:PCR-SSP(sequencespecificprimers-PCR)methodwasusedinstudy,thenwecalculatedthegenefrequencyofeachtype(directcalculationmethod:genefrequency=positivenumber/samplemultipliedby2)inordertocompareandanalyzewhetherornotsignificancebetweencontrolgroupandasthma.ThelevelsofIL-4,IFN-γweredetectedbyenzyme-linkedimmunosorbentassay(ELISA)inserum.3.Statisticalanalysis:statisticalanalysiswaspreformedusingthestatisticalprogramsSPSS17.0,thegenefrequencyofcontrolgroupandasthmagroupcanbecomparedbyChi-squaretest,OR(oddsratio),andconfidenceinterval,sizeoftestis0.05(bilateral).Pvaluelessthan0.05isconsideredtobestatisticalsignificance.TheconcentrationofIL-4andIFN-γwereexpressedasaverage±standarddeviation.ThecomparisonsamonggroupsusesLSD-test.ThecorrelationanalysisbetweenIFN-γandIL-4inasthmagroupwasanalyzedwithSpearmananalysis.Pvaluelessthan0.05isconsideredtobestatisticalsignificance.Results:(1)ThefrequencyofHLA-DQA1*0301intheasthmaticchildren20%wassignificantlyhigherthanthatinthehealthychildren(8.33%),χ2was7.159,Pwas0.007,ORwas3.333(95%confidenceintervalwas1.35-8.229).andthefrequencyofHLA-DQA1*0104inasthmaticchildren(27.78%)waslowerthanthatoftheinthehealthychildren(38.33%),χ2was5.234,Pwaslessthan0.05,ORwas0.380(95%confidenceintervalwas0.064-0.880).(2)Inasthmaticgroup,IL-4levelwassignificantl
