Cypate-loaded polymeric micellesforsimultaneous cancerimagingandefficientphotothermaltherapyAbstractCypate-loadedpolymericmicellesforsimultaneouscancer imagingandefficient photothermaltherapyAbstractObjective:Inthiswork,thepreparationandapplicationofcypate-loadedmicelles wereexplored.WedemonstrateatheranosticmicellesystemforcancerNIRFimagingwith enhancedsignaltonoiseratio(SNR)andsuperiorphotothermaltherapy.Thecopolymers consistingofmonomethoxypoly(ethyleneglycol)andalkylamine-graftedpoly(L-aspartic acid)areassembledwithcarbocyaninedye(cypate)intothetheranosticmicelles,which exhibitsmallsize,highloadingcapacity,goodstability,sustainedreleasebehavior,and negligiblecytotoxicity.Moreover,wefurtherobservethatthemicellesexhibitsuperior highimagingcontrastandphotothermaleffectoncancercells,causingefficient photothermaltumorregress uponphoto-irradiation.Methods:(1)Alkylamines(decylamine)wereconjugatedto theaspartateside chainsoftheblockcopolymer(poly(ethyleneglycol)-poly(aspartate)blockcopolymers) byamidebond,allthepolymerswerecharacterizedbyIR,1H-NMR.(2)Thepolymeric micellescontainingcypatewerepreparedbythedialysisandfilm-formingmethod, respectively. The Preparation procedures were optimized with the particle size,polydispersityindex（PDI）,entrapmentefficiency,drug-loading.Themorphologyand sizeofmicellesweremeasuredusingscanningelectronmicroscopy(SEM)anddynamiclight scattering(DLS),Thedrugloadingandencapsulationefficiencyweremeasuredusing UV-VISspectrophotometer,Thepyrenefluorescenceprobewasusedtoestimatethe criticalmicelleconcentration(CMC)ofmicelles.(3)MTTAssaywasusedtoestimatethe invitrocytotoxicityof copolymers onA549cells,Furermore,MTTassaywasalsousedto determinethephotothermalcytotoxicityofthemicellesagainstA549cellesunder785nm photoirradiationat1W/cm2.(4)Invitrocelluptakeofmicelleswasmeasuredusinga UV-VISspectrometer.(5)Wemonitoredthegenerationofsingletoxygen fromcypate using1,3-diphenylisobenzofuran(DPBF)asaprobewhichcanspecificallybequenched inthepresenceo
