PRODUCITONOFTHAILANDEPSINAFROMBURKHOLDERIATHAILANDENSISE264ABSTRACTRareBurkholderiathailandensisE264wasisolatedfromthericefieldofThailand.ItcanproduceThailandepsinAwhichcontainsabicyclicpeptidestructure.ThailandepinsAisaclassofhistonedeacetylaseinhibitors(HDACI)andastructuralanalogueofanti-cancerdrugsFK228withsignificantroleinT-celllymphomaandskincancertherapy.Currently，ThailandepsinAyieldproducedbywild-typestrainisverylow(10mg/L)andisnotsuitableforlargescalecommercialapplications.ThispaperattemptstoemployfermentaitionmediumoptimizationandmacroporousresinHP-20situextractionmethodstoimproveThailandepinsAproduction.Inthisstudy,Weprimarilyscreenedouttheproperageofseed,liquidvolume(ml),inoculum(%),temperature,pHandrotaryspeedusingone-factor-at-a-timescreening.Themediumcomponents,includingglucose,tryptone,phosphateandsodiumIIIcitratehadsignificanteffectsonThailandespinAproduction.ItiseffectivetochangeThailandepsinAdistributioninthefermentationbrothbyaddingHP-20resininbroth.TheinsituextractioneffectofmacroporousresinsonThailandespinAdecreasedthenegativefeedbackinhibitionandprovidedasimplerseparationstep．TheresultsshowedthatthroughadditionofmaeroporousresinHP-20，theyieldofThailandepsinAwassignificantlyincreasedby80%．Itgaveapreferableyieldwhenadditiveamountofresinis4%(w/v),addtimeis12handextractliquorischloroform.StatisticsbasedexperimentaldesignswereappliedtooptimizetheculturemediumcomponentsforenhancingThailandepsinAproductionbyBurkholderiathailandensisE264.Basedona24-1fractionalfactorialdesignandacentralcompositedesign,anoptimumconcentrationofglucose17.89g/L,typtone34.98g/L,K2HPO420.44g/L,KH2PO44.4g/Landsodiumcitrate0.01g/Lwasobtainedbyresponsesurfaceanalysis.ThepredictedmaximumThailandepsinAyieldwasashighas251.9mg/L,andwasfurtherverifiedbyvalidationexperiments.236.7mg/LThailandespinA,wasalmost2foldcomparedto104mg/LIVThailandepsinAfromonefactoratatimeoptimization.ThailandespinAfermentationkineticswasalsoinvestigated.KineticmodelsbasedonthemodifiedLogisticandLuedeking-Pir
