脱钙模拟椎体骨质疏松体外模型的建立
脱钙模拟evelope a model to mimic vertebral osteoporosis in vitro by demineralization
程自申1张智海2 刘莉3 尹磊4王岩 肖嵩华张永刚 张雪松摘要 目的 利用乙二胺四乙酸二钠盐(EDTA-Na2)体外脱钙模拟椎体骨质疏松,测量猪3联椎体骨密度和生物力学的特性,探讨一种短期制造骨质疏松性椎体压缩骨折模型的方法。方法取4个新鲜猪胸腰椎3联体,随机分成组,A.B.C.D.E.F.G每组7个标本,然后把B.C.D.E.F标本侵入0.4916mmol/L的EDTA-Na2脱钙液体中,分别在0、7、9、11、13、15天A.B.C.D.E.F.G骨密度、生物力学Mcro-ct检查。结果随着脱钙时间延长标本骨小梁变细、间隙增宽骨密度逐渐减低的趋势各组脱钙前后骨密度变化明显的差异性(p=0.0000)。随着脱钙时间的延长强度逐渐脱钙前后均有差异(p=0.0000)脱钙13天骨矿物质密度、组织骨密度、骨体积分数、结构模型指数、欧拉数、骨小梁连结密度均有明显的变化(p≤0.02)。结论采用0.4916mmol/L EDTA-Na2脱钙11天可模拟骨质疏松性椎体压缩骨折,是一种快速、可行、重复性高的方法。关键词:骨质疏松 EDTA-Na2; 原位脱钙 椎体压缩性骨折 生物力学Abstract: [Objective] Develope a short-term manufacturing the osteoporotic model to mimic the bioechanical characteristics of thoracolumbar osteoporosis utilizing the demineralization EDTA-Na2 (ethylenediaminetetraacetic acid disodium salt)to alter bone mineral density in In vitro. [Methods] 49 fresh porcine Thoracolumbar -3 -conjoined were randomly divided into seven groups, marked A.B.C.D.E.F.G,each seven specimens, and then all except for A were invasive into the EDTA-Na2 of 0.4916mmol / L, bone mineral density and biomechanical properties and Micro-ct inspection and of A.B.C.D.E.F.G were measured respectively and one out of each group was cut open at the time of the 0,5,7,9?,11,13,15 day,.?[Results] With the decalcification, general observation shows that bone trabecula of each group became thinner and the gap between bone trabecula became wider gradually ,the bone mineral density test show a trend that reduced, and the difference of BMD measurement bwteen pre- and post-demineraling process were statistically significant(p = 0.0000) ,and compression strength decreased, compared to compressive strength without decalcification, there were significant differences between pre- and post-demineraling .?After demineraling for 13 days Micro-CT detection showed that BMD?,TMD?, BVF SMI?,Euler Number Connectivity Density were significantly different compared to pre-demineraling (p ≤ 0.02)
