Ab Abstract PAGE PAGE IV ABSTRACT Objective Gap junctions (gap junction, GJ) played an important role on vasomotion, especially in regulating smooth muscle, and gap junction protein (Cx) is the base of gap junctions play function.Our team has been committed to research for Cx43 in cerebral vasospasm after subarachnoid hemorrhage (SAH) (CVS) , early research has shown that Cx43 mainly regulated vasoconstriction involved in the progress of CVS after SAH. But the CVS of the imaging and delayed ischemic neurological dysfunction (DIND) clinical symptoms is not completely consistent, according to the report of Cx43 existed widely in microvascular, this study assumes microvascular spasm after SAH is closely related to the expression of Cx43 in cerebral microvascular.To verify the hypothesis, the experiment used the secondary injection of blood SAH model in rats, to research whether the expression of Cx43 involved in the regulation of microvascular spasm after SAH and further understand its mechanism of action. Methods The induction of subarachnoid hemorrhage using a double blood injection, the double-hemorrhage model, was caused by repeating a second blood injection 24 h after the first induction of subarachnoid hemorrhage using the same method. SD rats model group: normal group, sham group, SAH3 day, 7 days group and adenovirus pretreatment SAH3 day, 7 days group. Using western blot, mmunohist- ochemical detection of each branch of middle cerebral artery microvascular(Diameter 20~80um) Cx43 expression level change;HE staining were observed the contraction of middle cerebral artery branchs、Inside diameter changes and smooth muscle cells and relevant pathological changes. Living fluorescence microscope detection: model group same as above, through the tail vein injection of FITC - dextran (dyed platelets), and used living fluorescence microscope detection the diameter of the arterial blood vessels in the PAGE PAGE V brain changes, comparison and analysis between
