AbstractObjectiveTostudytheinfluenceofrecombinantadenovirusvectoronproliferation andtheabilityofPRLsecretinginGH3cellsafterinhibitingtheexpressionofBMP-4, oberservewhetherGH3cellsaresensitivetorecombinantadenovirus,whichwilloffer experimentalfoundationandtheoreticalbasisforgenetherapyinthecontrolofhuman pituitary adenoma cells on the growth, invasionand the application ofhormone secretion.Methods(1)Designedandconstructedrecombinantadenovirusvectortargeting BMP-4shRNA.(2)GH3cellswereinfectedbyrecombinantadenovirusshRNA (rAd5-EGFP-BMP41+2+3+4shRNA)targetingBMP4andcarryingwithEGFPgene.The genesilencingefficiencyofAd5-BMP-4shRNAwasmonitoredbyRT-PCRandWesternBlot in GH3 cells．(3)The variation ofthe expression level of EGFRprotein were detectedbyWesternBlotrespectivelyafterGH3cellswereinfectedbytherAd5EGFP-BMP4 1+2+3+4shRNA. (4)AfterGH3cellswereinfectedbyrAd5EGFP-BMP41+2+3+4 shRNAthechangeofcellproliferationwastestedbytheMTSkit.(5)ELISAkitwas usedtodetectthevariationofthelevelofPRLsecretionafterrAd5EGFP-BMP41+2+3+4 shRNAinfected GH3 cells successfuliy.Results(1)RecombinantadenovirustargetedBMP4genewhichwasnamedrAd5 EGFP-1+2+3+4shRNABMP4wassuccessfullydesignedandconstructed,andtransfected packagingcellssuccessfullywithhighdegreeofrecombinantadenovirus.(2) BMP4-mRNAandproteinweresignificantlyinhibitedafterGH3cellswereinfectedby recombinantadenovirusrAd5EGFP-BMP41+2+3+4shRNA.(3)Theexpressionof EGFRproteinalsodowngradedafterBMP4genewasinterfered.(4)Aftersuccessfully infectedbyRAd5EGFP-BMP41+2+3+4shRNAtheODvaluewhichwastestedbythe MTSkitrisedmoreslowlythanthecontrolgroupovertime.(5)ThesecretionofPRL reducedafterGH3cellshadbeeninfectedbyrAd5EGFP-BMP41+2+3+4shRNAthan relative to the control group.ConclusionRecombinantadenoviruscarryingEGFPgeneandtargetingBMP4 shRNAissusceptibletoGH3cells.AsBMP-4genewassilencedthesecretionofPRLin GH3 cells were inhibitted,meanwhile,the proliferation of the GH3 cells can bedown-regulated.So BMP-4 can be used as one target of gene therapy
