Institute of Biotechnology, Shanxi University, Taiyuan 030006, China Corresponding author: Niu Lixi, Email: nlx@sxu.edu.cn Abstract Objective To observe the tumorigenic process, two pulmonary metastatic models of melanoma B16 cells were established and compared using C57BL/6 and KM mice, respectively. Methods Tail intravenous injection of 1×106 melanoma B16 cells/mouse resulted in macroscopic disease by 4-9 days and a randomly selected mouse was sacrificed in a certain interval to survey the morphologic characteristics of melanoma. The number of tumor nodules in the lung surface was counted and the size of tumor nodules was measured. The tumor-burdened time of mice was recorded and the tumor sections were made for pathological observation and analysis. Results HE staining showed that each mouse in the B16 cell injection groups had nest-shape metastatic foci distributed around the bronchi in the lung, but the tumor-burdened time was significantly different which was longer in KM mice than in C57BL/6 mice. Conclusion By comparing the characteristics of two pulmonary metastatic models of C57BL/6 mice and KM mice, we can concluded that the B16 melanoma models in KM mice were ideal tumor models which were suitable for researching the new drugs in lung cancer treatment.
