The application and clinical significance of fluorescence in situ hybridization and tissue array combined using in the early diagnosis of prostate cacinoma ABSTRACT Objective To investigate the exprssion of TMPRSS2-ERG and P504s in prostate and their cliniccal significance. Methods A tissue array containing 63 cases of various prostate diseases (30 cases of prostate carcinoma, 23 cases of benign prostatic hyperplasia and 10 cases of high grade prostatic intraepithelial neoplasia(HGPIN)) were analyzed by immunohistochemistry and fluorescence in situ hybridization (FISH) . The expression levels of TMPRSS2-ERG and P504s were compared between hyperplastic and cancerous prostate group. Results The expression of TMPRSS2-ERG gene fusion and P504s was high in prostate carcinoma but low in benign prostatic hyperplasia tissues, TMPRSS2-ERG and P504s expressed strongly in 70% and 86.7% cases of prostate carcinoma tissues, while slightly expressed in benign prostatic hyperplasia. TMPRSS2-ERG gene fusion was stably highly expressed in prostate carcinoma, and the expression was no related with the degree of prostate-specific antigen, Gleason score and TNM (p0.05), however, P504s - 2 - contrary (p0.05). Coincidence rate of TMPRSS2-ERG and P504s was enhanced with the increase of signal in P504s, and the negative coincidence rate was 66.7%.
