AbstractBio—effectsofsurvivalandetchingdamageoncellsurfaceandDNA strandbreaks
wereinvestigatedintheyeastsaccharomycescerevisiaeafterexposurebynitrogenionwithanen—
ergYbelow 40keV.Theresultshowedthat16%oftrehaloseprovideddefiniteprotectionforcells
againstvacuum stresscomparedwithglycero1.Incontrasttovacuum control,significantmorpho—
logicaldamageandDNA strandbreakswereobserved.inyeastcellsbombardedwithlow—energy
nitrogen,byscanningelectronmicroscopy (SEM)andterminaldeoxynucleotidyltransferase—
mediatedduTPnickendlabeling(TuNEL)immunofluorescenceassays.Moreover,PI(propidium
iodide1fluorescentstainingindicatedthatcellintegritycouldbedestroyedbyionirradiation.Cell
damageeventually affected cellviabilityandfreeradicalswereinvolved in celldamageasshown
byDMsO (dimethyIsulfoxide)rescueexperiment.Ourprimaryexperimentsdemonstratedthat
yeastcellscan beused asan optionalexperimenta1modeltostudythebiologicaleffectsoflow
energyionsandbeappliedtofurtherinvestigatethemechanism(s)underlyingthebio—effectsof
eukaryoticcells.
