· 510 · ORIGINALARTICLE ResearchontheRelationshipbetweenThickGreasyTongue FurFormationandVascularEndothelialCellPermeabilitywith theProteinExpressionofZonulaOccludens-1 QIWen—jie(齐文杰)，ZHANGMiao—miao(张苗苗)，WANGHong(王红)， WENYan(文艳)，WANGBao-en(王宝恩)，andZHANGShuw·en(张淑文) ABSTRACT Objective：TodeterminetherelationshipofthickgreasytonguefMrformationandpermeabilityof vascularendothelialcells(ECs)withtheproteinexpressionofzonulaoccludens-1(ZO-1)．Methods：Sprague DawleyratswererandomlydividedintoamodeIgroupofsevereacutepancreatitis(SAP)andasham·operated (SO)group．TheSAPratswerefurtherdividedintotwosubgroupsonthebasisoftongue。coatingstatus：a thickgreasytonguefurgroup(SAP—TGF)andanormaltonguefurgroup(SAP-NF)．Sixratswerechosen randomlyfr0m everygroupmentionedaboveforanEvansblueassay5daysaftermodelestablishment．F0r thehistomorphologyanalysis．theexpressionsofZO-1proteinandmRNAwerestudiedbyhematoxylin-eosin (HE)staining，transmissionelectronmicroscope，Westernblot，andQ—PCRusingbloodandtonguetissues， whichwerecollectedfrom 8ratsrandomlychosenfrom eachgroup．Results：Thepapillaedensityoftherat tonguesurfaceandthecaryocinesisfrequencyofthebasallayerweresignificantlyincreasedintheSAP-TGF groupcomparedwiththeSOgroup(P0．05)．EvansbluelevelsinthetonguetissueoftheSAP—TGFgroup weresignificantlyhigherthanthatoftheSOandSAP-NFgroups(P0．05)．VascularECswerewiderand obviouslyfissuredintheSAP-TGFgroupundertransmissionelectronmicroscopeobservation．Theproteinand mRNAexpressionofZO-1intheSAP．TGFgroupwerelowerthanthoseintheSAP-NF(P0．05)．Conclusions： Reproductiveactivityenhancementofglossalepithelialcellswasoneofthemaincharacteristicsofthickgreasy tonguefMrformation．Anincreaseinvasopermeabilitywascloselyassociatedwiththickgreasytonguefur formation．TightjunctionstructuraIvariationofvascularECsmightplayanimportantroleinthepathologicaland phys
