 纺锤体检测点蛋白 BUB1B 在胃癌组织中的 表达及意义# 墨青青，吴鹏，冯玲** 5 10 15 20 25 30 35 40 （华中科技大学附属同济医院妇产科，武汉 430030） 摘要：目的 检测胃癌组织中纺锤体检测点蛋白 BUB1B 的表达，探究 BUB1B 在胃癌发生中的 重要意义。方法 （1）采用荧光实时定量 PCR 检测 30 例胃癌组织及 17 例癌旁正常组织中 BUB1B mRNA 水平表达。（2）免疫组织化学检测 BUB1B 在胃癌组织及癌旁组织中 BUB1B 蛋 白的表达情况。结果 （1）在 mRNA 水平，BUB1B 在胃癌组织中的表达明显高于癌旁正常组 织，近 8.875±1.08 倍。（2）免疫组织化学结果显示，胃癌组织中 BUB1B 的表达明显高于 癌旁正常组织。按照评分标准统计显示，癌旁组织 BUB1B 表达均值为 92.12±12.99，胃癌 组织表达均值为 133.39±8.49，两者之间存在明显差异（p0.05）。且 BUB1B 表达与 TNM 分期相关，但与淋巴结转移，年龄，肿瘤大小及有无家族史无明显相关。结论 BUB1B 的过 表达在胃癌发生发展中起重要作用，为探索肿瘤发生机制及寻找更为有效的治疗提供新的思 路。 关键词：纺锤体检测点；BUBIB； 胃癌 Expression and significance of spindle checkpoint protein BUB1B in gastric cancer Mo Qingqing, Wu Peng, Feng Ling (Department of Obstetrics and Gynecology,Tong j i Hospital , Hua zhong University of Science and Technology , WuHan 430030) Abstract: Objective To study the expression of sp indle checkpoint p rotein BUB1B and explore the significant value of BUB1B in the initiation of gastric cancer. Methods (1)The mRNA expression levels of BUB1B were compared in tumor tissues of 30 gastric cancer patients with 17 corresponding normal tissues by real-time quantitative PCR. (2) The Immunohistochemistry was used to evaluate of BUB1B protein expression. Result (1) BUB1B mRNA expression levels were higher in gastric cancer than in corresponding normal tissues, neary 8.875±1.08. (2) The result of Immunohistochemistry showed , BUB1B protein expression were higher in gastric cancer than in corresponding normal tissues, according to the standard scoring , the mean BUB1B protein expression of corresponding normal tissue was 92.12±12.99 and the mean expression of gastric cancer was 133.39±8.49 (p0.05). And the expression of BUB1B is evidently related with TNM stage, but there was no evident correlation with lymph nodes metastasis, age, tumor size and family history. Conclusion The overexpression of BUB1B is related with the progression of gastric cancer and provides an new thread to explore the mechanism of cancer initiation a
