GGT 调节 HIF-lα 对胶质瘤侵袭作用的研究中文摘要目的研究低轼条件 1蛋白异戊烯转移酶 GGT 对脑胶质瘤 U251 细胞侵袋和迁移能力的影响及机制，为胶质瘤的基因泊疗提供理论和实验依据。方法(1)用氧化铀 (CoCh) 处理细胞得到化学性低轼模型，用免疫印地法 (WB)检测 HIF-lα 水平以明确低氧模型是否成功:划痕实验成 Transwell 实验检测细胞迁移侵袭能力:明胶酶谱法检测 MMP2 fJ击性水平:细胞贴哇实验阳则细胞拈附能力; WB 险测 Cadherin 水平。 (2) WB 和免疫荧 :Jt法检测低氧条件下 GGT特异性 P 哑塔的水平。(3)低轼条件 F ，用siRNA r调 GGT? 表达，或用。GT特异性抑制剂 GGTI-2147 处理脑胶质瘤 U251 细胞， WB 检测 GGT? 和 HIF-lα 的水平。 (4)低氧条件 f ，下调GGT? 表达和 GG丁1-2147 处理后， Transwell 实验检测脑胶质瘤 U251 细胞的侵褒迁移能力。钳果 (1 )化学性低氧条件下，脑胶质瘤 U251 细胞佼袭迁移能力降低，粘附能力升高。 (2)化学性低氧条件下， GG丁P 水平逐渐升高，在低氧 24h 达高峰。(3) 化学性低氧条件下， GGT 表达下调成活性抑制均使 HIF-la 表达下降。 (4) 化学性低氧条件下， GGT 表达下调戒活性抑制后，脑胶质瘤 U251 细胞的迁移和侵褒能力下降。、结论(1 )在氧化钻低氧模型中，脑胶质瘤 U251 细胞的迁移和侵袭能力均下降。 (2) 低氧条件下 GGT 对 HIF-la 有调节作用。 (3) 化学性低氧条件下， GGTk可能通过调节日IF翩 1α 调控脑胶质榴细胞的侵袭和迁移能力。关键词脑股质捕; GGT;HIF斗 α; 侵袭;迁移GGTase regulates migration and invasion of glioma cells through HIF斗αactivationAbstractObjective To study the role of GGT in the migratory and invasive ability of glioma cel1s and to investigate the possible mechanisms.Methods 1，A伽 CoCh treatment (the chemical hypoxia statement)，the protein level of HIF-lαwas examined by westem blotting.ηle migratory and invasive abilitywere tested by wound healing assay and transwelI invasion assay respectively.
