Nitric oxidesuspensionoferythrocyte deformabilitymechanismAbstract:Nitric oxide (NO) in vivo as a highly reactive free radicals, with a wide range of physiological effects. NO through a combination of red blood cell hemoglobin (Hb) in the conservative β93Cys residues (Hbβ93Cys), to form S-nitroso-hemoglobin (SNO-Hb). Hb molecule NO from the heme transfer to Hbβ93Cys thiol regulation of NO storage and release, mediated vasodilatation. Library of red blood cells with the extension of storage time, NO massive loss, resulting in red blood cell vasodilator activity, erythrocyte deformability capacity and hemoglobin oxygen carrying capacity of physiological activity. Therefore, the understanding of NO in the loss of red blood cell storage process, understanding the mechanism of action of NO in the circulatory system, red blood cell transfusions for the protection of safety and effectiveness of the significance and value. This study by adding NO to improve deposit red blood cell deformability, explore the mechanism of NO on red blood cell morphology, with theoretical and practical significance for the further evaluation of the clinical transfusion.Objective: this experiment by adding reagents of different concentrations of NO: L-Arginine (L-arginine / NO precursor), to determine the suspension of red blood cells red blood cells of different storage phasedistortion indicators to explore NO and banked blood red blood cell deformability mechanism to further assessment of storage quality of the suspension of red blood cells stored.Methods: 10 cases of fresh suspension of each 1u red blood cells, each case was distributed into 4mL / tube, a total of 500, grouping number, storage, dedicated storage in the refrigerator.
