AbstractAsthesubstrateofthebiosensorfabrication,thebiosensinginterfaceitselfcouldplayaroleofmodificationplatformforthebiosensingprobeandaffordafriendlyatmosphereforthebiochemicalreactionbetweentheprobemoleculeandtargetmoleculeaswellastheinteractionbetweenprobes.Thus,itisameaningfulandsignificantworktobuildaneffectivebiosensinginterface,optimizetheproberecognitioneffectivenessandthebiosensorperformanceformaximumefficiency,andultimatelyrealizethesensitivitypotentialdevelopment.Withthetargetofbuildinganultra-sensitivebiosensorinterfacewithfinenanostructure,thepaperputforwardtwonovelideasofassemblystrategy:nanogoldparticlearrayformattingmethodandsinglemoleculeprobefabricationmethodbasedonthree-dimensionalporousgraphenemodification.ThepollutingionofHg2+ischosentobethetargetmodeltoconstructkindofelectrochemiluminescencebiosensorandelectrochemistrybiosensor.Basedonthepioneer’sstudyresultoftheeffectofoptimizedDNAmoleculedistributionandprobestrandsorientationforimprovingDNAhybridizationefficiency,wedesignedapromisingDNAfabricatingapproachapplyingthenumberdensitycontrolledGNPsarrayasself-assemblyplatformtoruletheDNAstrandsdistributionandenhancetheproberecognitionefficiency.Withtheelectrochemicaldepositionmethod,chloroauricacidasdepositionsolutionwasusedtobuildagoldnanoparticlepatternedglasscarbonelectrodesurface,meanwhilethearraynumberdensityanddotdiameter(~50-60nm)canbecontrolledbyadjustthesolutiondensityandelectrochemicaldepositiontime.Withthemodificationplatformindifferentarraynumberdensity,ahairpin-DNAlabeledwithferroceneasECLquencherandanothershortDNAstrandlabeledwithRu(bpy)32+asreporterwerehybridizedtoconstructthesignalgeneratorinoff-state.BasedontheT-Tmismatchrecognitionstrategy,thehairpinDNAwithanotherthymine(T)-richcapture-DNAwasintroducedtoformT-Tmismatcheswiththeloopsequenceofthehairpin-DNAinthepresenceofHg2+andinducethestem-loopopen,meanwhiletheECL“signal-on”wastriggered.Thepeaksensitivitywiththelowestdetectionlimitof0.1nMwasachievedwiththeoptimalGNPsnumberdensity
