ExpressionandSignificanceofBmi-1GeneinPapillary thyroid carcinoma AbstractBackground:Bmi-1（B-cellspecificmoloneyleukemiavirusinsertionsite1）,one importantmemberofPCG(Polycombgroupgenes)family,wasdiscoverbyHollandCancerCentre from mice,slymphomain1991,itplaysanimportantroleinformationofmanymalignanttumors.Manyresearchhaveprovethatasanoncogene,Bmi-1hadparticipatedinmanyhumanmalignanttumors, such as leukemia【1】、breast cancer【2】，small cell lung cancer【3】andgastrointestinalcarcinomas【4】.Ontheotherhand,fromaspectoftumormetastasis,BruggemanSWhaveprovedthatBmi-1canenhancetheadhesionbetweentumorcellsandextracellularmatrixthroughintegrinreceptors.ThereisnoresearchabouttheexpressionofBmi-1inthyroidcarcinoma ,hitherto. ObjectiveToanalyzetheexpressionofBmi-1geneinthetissueofthyroiddiseasesandprobetherelationshipbetweentheBmi-1andpathogenesisof the papillary thyroid carcinoma and its lymph metastasis. Materials: 1. AllthethyroidspecimenswerecollectedfromTheSecondGeneral SurgeryDepartmentofGuangzhouFirstMunicipalPeople’sHospitalbetween June,2008 to June 2009. 2. The reagents about extracting RNA and two-step RT-PCR Kit Methods:Using18casesofnodulargoitersand18normalthyroidtissuesasthecontrolgroup,while24papillarythyroidcarcinomaasexperimentalgroup,usingtwo-stepRT-PCRtodetectexpressionofBmi-1between normalthyroidtissue、nodulargoiterandpapillarythyroid carcinoma,thenusingR×Csplitχ2testtoanalyzetheresultanddetermine the expression of Bmi-1 between the three groups.Result:1.From24papillarythyroidcarcinoma,23casesBmi-1positive, positiverateis95.83%；From18nodulargoiters,only2casesBmi-1positive,positiverateis11.11%；Bmi-1expressionarenegativeinallnormalthyroidtissues.ThereisasignificantdifferenceofBmi-1 expressionbetweenpapillarythyroidcarcinomagroupandnodulargoitersgroup（χ2=30.643，p0.05）；andthereisalsoasignificantdifferencebetweenpapillarythyroidcarcinomagroupandnormalthyroidtissuegroup（χ2=38.131，p0.05）；butnosignificantdifferencebetweennormalthyroidtissuegroupandnodulargo
