ABSTRACTFromthemid-twentiethcentury,thegold-containingdrugshavealreadybeenusedforthetreatmentofrheumatoidarthritis.Alarge numberofAu(I)andAu(III)complexesweresuccessfullysynthesized,amongwhichthemostfamousforAu(I)classisAuranofin.Thesedrugsare mainlyusedforthetreatmentofactiverheumatoidarthritis.Theycandelaytherheumatoidarthritislesions,improvesymptoms,andhavegoodtolerance.Au(III)andPt(II)haveidentical4f145d86d0closed-shellelectronconfiguration,andtheircoordination modestake thesamesquareplanarconfiguration.InasmuchasthePtanti-cancerdrugsproducea seriesofsideeffects, alargenumberofAu(III)classofgoldcomplexesaresynthesizedasanewpotentialanti-cancerdrugs.Andtheircytotoxicityhas become arecentresearchfocus.Au(III)complexesaspotentialanti-cancerdrugshowamorevariablechemicalconfiguration.Theconfigurationsofinterestreportedintheliteraturemaybesummarizedintofourcategories:(i)thesingleligand;(ii)coordinatedbyporphyrin;(iii)coordinatedbyavarietyoforganicmolecules;(iv)coordinatedbybi-dentateligands.Awide varietyofAu(III)complexesarereportedintheliterature,butmost ofthemfocusedonthestructuralcharacterizationandinvitro experiments.Inthefewinvivoexperiments,thechoiceoftumorcelllinesandexperimentalconditionsaredifferent,leadingtothecompounds’anti-canceractivitydonotshowregularityandcomparability.Sofarthe anticancermechanism of the Au(III)complexesare stillincontroversy,whichincludesthefollowingaspects：(i)Thestructure-activitymechanisms,includingtheimpactof takingoffgroups andtheinfluenceofligands.Easilylosingligands andaromaticconjugategroupsofAu(III)complexeshaveastrongcytotoxicity.(ii)thesulfur-containingorganicbio-molecularinteractions.SulfhydrylgroupsandthereactivityofAu(III)complexescanlimitthe progressionofthecancercellgrowth,ofwhichthebiologicaltargets maybe the thiol-sulfuratom.(iii)the interactionbetweenproteinsandtheirprotease.ThecenterofAu(III)complexmayhaveastrongcoordinationwithhistidine.(iv)theinteractionoftheDNAfragments.ForAu(III)hastheidenticalelectronicconfigura
