大孔吸附树脂分离纯化洋葱皮黄酮的研究-河南农业科学
※工艺技术 食品科学 2011, Vol. 32, No. 12 133 1 1, 2 1 1 徐怀德 ,陈 佳 * ,包 蓉 ,刘 坤 ,李 晋 (1.西北农林科技大学食品科学与工程学院,陕西 杨凌 712100 ;2.西北农林科技大学林学院,陕西 杨凌 712100) 以黄酮含量为指标,比较6 种大孔吸附树脂对洋葱皮黄酮的吸附和解吸效果。通过静态吸附与解吸实验, 筛选出效果较好的X-5 树脂进行动态实验研究。结果表明:X-5 树脂纯化洋葱皮黄酮的工艺条件为吸附流速2mL/min 、 上样溶液pH5.0 、上样液质量浓度0.5mg/mL 、50mL 80% 乙醇作为洗脱液,洗脱流速lmL/min 。经X-5 树脂纯化 后,洋葱皮黄酮纯度从 7.95% 提高到 80.78% 。三次重结晶后其纯度可达94.5% 。该方法简单可行,纯化效果好, 适合于工业化生产。 大孔吸附树脂;分离;纯化;洋葱皮;黄酮 Separation and Purification of Flavonoids from Onion Peel by Macroporous Resin Adsorption 1 1, 2 1 1 XU Huai-de ,CHEN Jia *,BAO Rong ,LIU Kun ,LI Jin (1. College of Forestry, Northwest AF University, Yangling 712100, China) Abstract :The characteristics of adsorption and desorption of onion peel flavonoids on six kinds of macroporous resins were compared. Static adsorption and desorption experiments indicated that X-5 resin was the best resin for the separation and purification of flavonoids from onion peel. The optimal process parameters for purifying onion peel flavonoids with X-5 resin were sample loading flow rate of 0.5 mg/mL, sample pH 5.0 and 50 mL of 80% ethanol of as eluent at a flow rate of 1.0 mL/min. After the purification, the purity of flavonoids from onion peel was increased from 7.95% to 80.78% and could reach up to 94.5% after further triple recrystallization.
