响应面法优化超声波辅助提取甘草多糖工艺的研究 张琳，樊金玲，朱文学，马海乐，范翠英 （河南科技大学食品与生物工程学院，河南 洛阳 471003） 摘 要：试验研究了乌拉尔甘草多糖的超声波辅助提取工艺。在单因素实验的基础上，选定超声功率、时间和提取温度三个因素的三个水平进行中心组合实验，建立甘草多糖率的二次回归方程，其决定系数为98.98%91.67%。通过响应面分析及岭嵴分析得到优化提取工艺条件为超声功率580W、时间64.5min、提取温度64.6℃该条件下多糖率预测值为9.62%，验证值为9.56%，此值为相同条件下传统水浴浸提法的三倍多预测值为%，验证值为%。红外光谱检测结果显示，超声波提取法与传统浸提法所得甘草粗多糖基本构成相同。关键词：乌拉尔甘草；多糖；超声波辅助提取；响应面分析；岭嵴分析；红外光谱检测 Optimization of the ultrasound-assisted extraction of polysaccharides from Glycyrrhiza uralensis Fish. using response surface analysis Zhang Lin ，Fan Jinling , Zhu Wenxue , Ma Haile , Fan Cuiying (College of Food and Bioengineering , Henan University of Science and Technology, Luoyang 471003, China) Abstract: A three-level Box-Behnken design, combined with the canonical and ridge analyses, was employed to optimise the process parameters for polysaccharides ultrasound-assisted extraction from the Glycyrrhiza uralensis Fish., one of the most valued traditional Chinese medicines. The critical factors selected for the investigation were actual sonic power, duration of time and extraction temperature. The experimental results were fitted with a second-order polynomial equation by a multiple regression analysis and more than 98.98% and 91.67% of the variation could be predicted by the yield and the purity of polysaccharides models. The canonical analysis of surface responses revealed that the three eigenvalues had different signs, indicating a saddle stationary surface. The optimal conditions for extraction of polysaccharides from the Glycyrrhiza uralensis Fish. were determined, using the ridge analysis, as extraction time, 64.5?min; extraction temperature, 64.6?°C; at actual sonic power of 580W. Under these conditions, the highest yield and the purity of polysaccharides reached with estimated value and verified value being of 9.62%、9.56% and 71.36%、65.65% , respectively. It is three times higher than the classical extraction yield and the structure of polysaccharides from the two methods are the same through IR. K
