宁夏医科大学硕士研究生学位论文  英文摘要 The Effect of Post Staged Sirolimus Drug-eluting Stents Implantation Dual Anti-platelet Therapies on Platelet Activation ABSTRACT Objective  The study investigated the changes of platelet fibrinogen receptor exrpression (PAC-1), P-selectin (CD62P), ThromboxaneB2 (TXB2), Endothlin (ET-1), Fibrinogen (FIB) in patients received staged coronary Sirolimus drug-eluting stents implantation. To evaluate th effect on platelets activation of aspirin and clopidogrell dual anti-platelet therapies. Methods  40 in-hospital patients with stable coronary heart disease(CHD) were enrolled in this study. All patients were diagnosed CHD (at least one steonsis epicardial artery lesion≥75%) by coronary angiography(CAG). Asprin and clopidogrel were taken preoperation no less than three days to effectively inhibit platelet activation by all patients. They were divided in to two groups:stent group (n=20) patients undergone staged Sirolimus drug-eluting stents implantation, CAG group (n=20) patients undergone CAG.. Peripheral artery blood samples were colleted before the procedures and after procedures immediately and at 6 hours, 24 hours. PAC-1 and CD62P were determined by flow cytometry (FCM), TXB2 and ET-1, FIB were analyzed with enzyme-linked immunosorbent assay(ELISA). Observing in-hospital and follow-up major adverse cardiovalcular events(MACE). Results After coronary stents implantation, expression of PAC-1 and CD62P significantly increased in contrast with pre-stents implantation,peaking at 6 hours (PAC-1， P=0.001), (CD62P，P=0.008), (PAC-1+CD62P，P=0.003), TXB2 significantly increased, peaking at 24 hours(P=0.002). Plasma levels ET-1 and FIB were no defference before and after stents implantation. After CAG, expression of PAC-1 and CD62P, level of TXB2, ET-1 and FIB were no difference contrast with pre-CAG, No MACE happened.
