SN50对大鼠视网膜缺血再灌注损伤的保护作用研究 [目的] 探讨NF- κB活性抑制剂SN50对大鼠视网膜缺血再灌注损伤的保护作用。[方法] 将成年雄性SD大鼠随机分为正常对照组、视网膜缺血再灌注组、视网膜缺血再灌注+SN50组,每组9只,选取在缺血再灌注后的6h、24h、72h三个时间点,通过HE观察大鼠视网膜的病理变化,免疫组织化学检测NF-κB的表达部位,实时定量PCR检测TNF-α的表达。[结果] 缺血再灌注6h后视网膜出现组织轻度水肿,少量细胞变性,随着时间的延长,视网膜的损伤逐渐加重,经过SN50注射处理后,视网膜的损伤明显减轻;缺血再灌注6h后视网膜上可见NF-κB的阳性表达,经过SN50注射处理后,NF-κB的阳性表达在缺血再灌注24 h后明显减弱;经过SN50注射处理后,与未注射SN50组比较,24h后视网膜的TNF-α表达已有统计学差异(p0.05)。[结论] SN50减轻了大鼠视网膜的病理损害,对大鼠视网膜缺血再灌注损伤中NF- κB 的表达具有抑制作用,而且还能影响TNF-α的表达,因此SN50对视网膜缺血再灌注损伤具有保护作用。 关键词:缺血再灌注,SN50,NF-κB,TNF-α,视网膜 Study on the protective effect of SN50 on retinal ischemia reperfusion injury in rats [目的] 探讨NF- κB活性抑制剂SN50对大鼠视网膜缺血再灌注损伤的保护作用。[方法] 将成年雄性SD大鼠随机分为正常对照组、视网膜缺血再灌注组、视网膜缺血再灌注+SN50组,每组9只,选取在缺血再灌注后的6h、24h、72h三个时间点,通过HE观察大鼠视网膜的病理变化,免疫组织化学检测NF-κB的表达部位,Real-time PCR检测TNF-α的表达。[结果] 缺血再灌注6h后视网膜出现组织轻度水肿,少量细胞变性,随着时间的延长,视网膜的损伤逐渐加重,经过SN50注射处理后,视网膜的损伤明显减轻;缺血再灌注6h后视网膜上可见NF-κB的阳性表达,经过SN50注射处理后,NF-κB的阳性表达在缺血再灌注24 h后明显减弱;经过SN50注射处理后,与未注射SN50组比较,24h后视网膜的TNF-α表达已有统计学差异(plt;0.05)。[结论] SN50减轻了大鼠视网膜的病理损害,对大鼠视网膜缺血再灌注损伤中NF- κB 的表达具有抑制作用,而且还能影响TNF-α的表达,因此SN50对视网膜缺血再灌注损伤具有保护作用。 [Objective] To investigate the protective effects of SN50, which is the NF- κB inhibitor on retinal ischemia reperfusion injury in rats. [Methods] SD rats were randomly divided into the normal control group, the retinal ischemia reperfusion group and the +SN50 retinal ischemia reperfusion group. Each group has 9 rats . Pathological changes of rat retina were observed at 6h ,24h and 72h after ischemia reperfusion by HE, NF- κB expression sites were detected by immunohistochemistry ,and the expression of TNF- α were detected by real-time PCR. [Results] 6h after ischemia reperfusion ,the retina tissue was edema, a small amount of cells were degeneration. With the extension of time, the retinal injury aggravated gradually. However, after injection SN50 after treatment, the retinal injury reduced significantly; The expression of NF- κB were observed in retinal 6h after ischemia reperfusion. The positive expression of N
