AbsttactCapillary Absttact Capillary electrochromatography(CEC)is a new miniaturized separation technique that combines the advantages of high performance liquid chromatography(HPLC)and capillary electrophoresis(CE)Due to high efficiency,high selectivity,rapid separation and low consumption of both samples and solvents.CEC has received considerable attention io recent years and evolved as a promising analytical technique.However,there were still problems and di币curies associated with bubble formation and column dry-out in CEC There problems can be addressed by pressurized CEC(PCEC)and capillary monolithic column.To data,the two techniques have been the most interesting fields in CEC.In this paper,research works on the these were done as the following. A special CEC system has been developed.By optimizing operation condition and instrument hardware,a continuous gradient elution can be carried out conveniently on this instrument.Six peptides can be separated successfully by gradient hydrophobic interaction-PCEC and gradient hydrophilic interaction-PCEC respectively.Gradient PCEC is powerful and useful for complicated peptides separation,which is difficult in isocratic elution mode. Separation of peptides was investigated on mixed mode of hydro)philic interaction and strong cation exchange PCEC first time.The effects of applied voltage,supplementary pressure,organic modifier concentration.ionic strength and pH value in the buffer oo PCEC separation were investigated.In addition.a mathematical model was developed to describe the relationship of capacity factor with experimental condition in PCEC.and the model equations were used to evaluate the retention behaviors of peptides. Separation of neutral,acidic and basic compounds were obtained.The base separation of six basic drugs was obtained using mixed mode PCEC first time.The effects of mobile phase conditions on retention behaviors of solutes were investigated.Moreover the effects of pressure and voltage on separation
