达特异性,且均在根中的表达量最高;GbVWRl、GbVWR2均可以对黄萎病菌和MeJA、 达特异性,且均在根中的表达量最高;GbVWRl、GbVWR2均可以对黄萎病菌和MeJA、 SA、ET和GA激素处理做出应答反应。GbVWRl蛋白定位在细胞质膜系统,GbVWR2 蛋白定位在细胞核。GbVWRl和GbVWR2均对棉花抗黄萎病反应具有正向调节作用。 关键词:海岛棉;抗黄萎病;未知基因;克隆;VIGS;功能分析 万方数据 Cloning Cloning and functional analysis of VWR genes from Gossypium barbadense in Verticilliam wilt resistance Author:Zhang Li-jia Major:Crop Genetics and Breeding Supervisor:Prof.WangXing-fen Prof.Ma Zhi-ying Abstract Verticillium wilt(Ⅵ聊,caused by the soil-borne fungus Verticillium dahliae站eb,is the most destructive disease in coRon production in China.Planting VW-resistant cultivars is the most cost-effective and economical control method.Gossypium hirsutum and Gossypium barbadense,belonging to the predominant cultivated species,originated from the same ancestor.However,tlley differ in many morphological and agronomic traits.G hirsutum is high yielding and widely adapted to different growing conditions or environments but generally susceptible,while G barbadense has superior extra-long, strong and fine fiber properties and hiigh resistance but witll lower yield potential and narrower adaption.Target transferring the VW-resistant trait or function genes from G barbadense to G hirsutum is an important strategy to improve coRon VW-resistance. Therefore,identification of disease-resistance genes and studying their function would be of great significance for coRon disease resistant breeding. In this study,we identified two genes that involved in coRon defense againstⅣ dahliae based on screening the full—length eDNA library and suppression subtractive hybridization library(SSH)induced by形dahliae in G barbadense and Gossypium hirsutum,respectively.Sequence analysis indicated that there were no any annotation in NCBI database,and we named the sequences from G barbadense as GbVWRl and GbVWR2.In this study,we characterized GbVWR genes and analyzed tlleir expression and function against形dahBae.The main results were as follows: 1.The GbVWRl and GbVWR2.including 1 9
