Studies have shown that the development of restenosis is related to intimal proliferation and vessel remodeling. The state of uncontrolled activation, migration and proliferation of vascular smooth muscle cell(VSMC) and the abnormal metabolism of extracellular matrix(ECM) both play important roles in restenosis. And the abnormal synthesis and degradation of collagen is one of the important reason of vessel remodeling. In the present study, we set up a model of vessel stenosis after balloon injuries in rabbits’ iliac arteries, emulating the pathology of restenosis. We investigated the effect Tetrandrine on the proliferation of VSMCs and collagen content of vessel wall, with the intention of finding out whether Tetrandrine can inhibit VSMCs and collagen synthesis in vivo. Methods Intima denudated model of rabbits was employed. 24 male New Zealand rabbits were randomized into three groups: Tetrandrine-treated group, control group and sham group. in TET group, Tetrandrine was administered 3 days before and until 28 days after 3 angioplasty(45mg·kg-1·d-1). The control group underwent the same balloon angioplasty while no medicine was administered. No intervention was carried out in the sham group. Target vessels were harvested at 28 days after angioplasty. Histomorphology study, including HE and Masson’s staining, was applied. Lumen area, intimal area, mean intimal thickness and medial area was measured by computed image analysis system.
