第四章：本文利用CdSe量子点的荧光增强现象建立了一种检测痕量人血红蛋白的新方法，并探讨了CdSe量子点荧光增强的机理。在最佳测试条件下，血红蛋白浓度在0.375～105ng·ml?1范围内，CdSe量子点的荧光增强与血红蛋白浓度呈良好的线性关系。检测限是0.18ng·ml?1，对于30ng·ml?1的血红蛋白而言该法的相对标准偏差为0.41%(测定9次)。该法的抗干扰性较强，用于测定血样中的血红蛋白结果令人满意。关键词：银纳米粒子，CdS量子点，CdSe量子点，血红蛋白，结合常数，相互作用，荧光猝灭TheInteractionbetweenHemoglobinandAg、CdSandCdSeNanoparticlesanditsbiologicalapplicationPostgraduate:LiuXinyanSupervisors:Prof.LIANGHong,Asso-Prof.SHENXingcanAbstractChapterone:Inthischapter,theapplicationofnanoparticlesforbiologicaltechnologyissummarized,includingtheapplicationfortheclinicaldiagnostics,DNAarraytechnology,anddirecteddeliveryofdrugs.Theprogressinhemoglobinanditsinteractionwithnanoparticles,suchasAg、CdS、andCdSenanoparticles,arereviewed.Lastly,weintroducetheresearchsuccessofourteamandthesignificanceofthisarticle.Chaptertwo:Inthispaper,theinteractionbetweenbovinehemoglobin(BHb)andAgnanoparticles(AgNPs)hasbeeninvestigatedbyultraviolet-visible(UV-Vis),fluorescence,synchronousfluorescence,circulardichroism(CD)andFouriertransforminfrared(FT-IR)spectroscopies.Thedecreaseandred-shiftof415-nmsurfaceplamonbandofAgNPsindicatesthattheBHbcanbeadsorbedonthesurfaceofAgNPs.TheSoretbanddecreasesgraduallywiththeincreasingamountofAgNPs,suggestingthatthedetachmentofsomehemechromophoresfromtheirmatrixesinBHb.ThefluorescenceintensityofBHbisquenchedbyAgNPs,andtheanalysisofStern-Volmerequationrevealsthatthemechanismisastaticquenchingprocedure.ThebindingconstantKwasobtainedwithcalculationofthespectradata,andtheorderofmagnitudeofKisfoundtobe109~1010.Theblue-shiftofsynchronousfluorescencespectralrevealsthatthemicroenvironmentsaroundtryptophanandtyrosineresiduesaredisturbedbyAgNPs,whichinducedtobeburiedinsidethehydrophobiccavities.Thecalculationoffar-UVCDdatashowthatthesecondarystructureofBHbhasslightchanges,andtheα-helicalcontentdecreases.Inaddition,theFT-IRspectraprovidetheevidencethatthesulphuratomsincysteineresidues,carboxyloxygen,andnitrogenatomsinpeptideorresiduesprobablyhavethedirectchemicalbondsonsurfaceofAgNPs.Chapterthree
