的影响研究 中文摘要
目的:观察白介素-33 抗体干预变应性鼻炎小鼠模型的造模过程,探讨其对鼻黏膜 炎症及基质金属蛋白酶-9 表达的影响。
方法:选取 BALB/c 小鼠(8-10 周)24 只,雌雄各半,分别随机分为三组,正常组 (n=8),模型组(n=8)和干预组(n=8)。采用卵清蛋白致敏法建立变应性鼻炎小鼠模 型。干预组在每次局部激发前腹腔注射 IL-33 单克隆抗体,模型组及正常组等量生理 盐水替代。应用酶联免疫吸附法检测各组小鼠外周血清中卵清蛋白特异性 IgE 浓 度;苏木素-伊红染色后显微镜下观察各组小鼠鼻黏膜组织形态及嗜酸性粒细胞浸润 情况;免疫组化法测定各组小鼠鼻黏膜中基质金属蛋白酶-9 的表达情况。
结果:1.白介素-33 抗体干预后变应性鼻炎小鼠抓挠及喷嚏次数明显减少。2. 干预组
中小鼠血清中卵清蛋白特异性 IgE 含量显著低于模型组,组间差别有统计学意义 (P0.05)。3. 苏木素-伊红染色结果示白介素-33 抗体能够减轻变应性鼻炎小鼠鼻黏膜 炎症并减少炎性细胞浸润。4.免疫组化结果显示白介素-33 抗体能降低基质金属蛋白 酶-9 在变应性鼻炎小鼠鼻黏膜中表达。
结论:1.通过卵清蛋白经典致敏方法可成功建立变应性鼻炎小鼠模型。2.白介素-33 抗体干预后能够减轻变应性鼻炎小鼠鼻部症状及嗜酸性粒细胞的浸润。3.白介素-33 抗体能够抑制基质金属蛋白酶-9 在鼻黏膜中表达从而抑制鼻黏膜结构破坏。

Investigation the effects of Interleukin-33 antibody on nasal mucosa inflammation and MMP-9 expression in mice with allergic rhinitis
Abstract
Objective: To investigate the effect of Interleukin-33(IL-33)antibody on nasal mucosa inflammation and matrix metalloproteinase-9(MMP-9) expression in mice nasal with allergic rhinitis.
Methods:Twenty-four BALB/c mice that 6 to 8 weeks were invovled. These mices were randomly divided into three groups. The normal control group (n = 8), model group (n = 8) and the intervention group (n = 8). Ovalbumin was used to established mice model of AR. The intervention group mices were treatmented with IL-33 antibody before local excitation, Model group and normal control group are use by the same amount saline replaced. Enzyme-linked immunosorbent assay (ELISA) to detect the concentration of OVA-specific IgE in serum of each group of mice. Under the microscope observed nasal morphology and eosinophil infiltration cases of each group of mice after HE staining. Each group of mice was measured expression of MMP-9 in nasal mucosa by immunohistochemistry(SP).
