第一部分Hoxa11调控子宫内膜容受性的动物研究摘要目的：通过宫腔内脂质体转染的方法，上调和下调小鼠围着床期子宫内膜Hoxa11的表达水平，探索Hoxa11，Integrin-β3，β-catenin，leukemiainhibitoryfactor(Lif)四种因子在基因和蛋白水平表达变化规律。方法：选择未生育过的性成熟昆明小鼠，分为两组（采用自身对照）：其中一组于妊娠第2天向每只小鼠“Y”形子宫两侧宫角内分别注入pEGFP-Hoxa11表达质粒和空白质粒，另一组于妊娠第2天向每只小鼠两侧宫角内分别注入Hoxa11的SiRNA表达质粒和空白质粒。于妊娠第5天处死小鼠，剖取其子宫组织，通过RealtimePCR和Westernblot技术分别从基因水平和蛋白水平筛选出Hoxa11转染成功的小鼠，再分别从基因和蛋白水平检测Integrin-β3，β-catenin，leukemiainhibitoryfactor(Lif)三个因子在两组小鼠子宫内膜上的表达差异。结果：和空白质粒小鼠相比，Hoxa11上调和下调成功的子宫内膜中，Integrin-β3，β-catenin，leukemiainhibitoryfactor(Lif)三个因子呈现表达明显上升和下降趋势，与Hoxa11变化趋势一致。结论：四种因子在调控子宫内膜容受性方面是互相关联的；Hoxa11可能通过改变着床期内膜Integrin-β3，β-catenin，Lif等分泌因子的表达来影响子宫内膜容受性。关键词：Hoxa11;Integrin-β3；β-catenin；Lif；上调；下调；内膜容受性PartITheanimalresearchofHoxa11regulatingendometrialreceptivityABSTRACTObjective:ToexploretheexpressionrelationshipofHoxa11，Integrin-β3，β-catenin，leukemiainhibitoryfactor(Lif)onthegeneandproteinlevel,up-regulateanddown-regulatetheexpressionofHoxa11inmiceuterithroughtransfectedwithplasmid-liposomecomplexes.Methods:SelectnulliparoussexualmaturityKunmingmicedividedintotwogroups(Usingself-control):ThefirstgroupwereinjectedthepEGFP-Hoxa11andemptyplasmid-liposomecomplexesintobothsidesofY-shapeduterusofpregnancyD2micerespectively;theothergroupwereinjectedwithHoxa11SiRNAandemptyones.ExcisetheuteritissueafterthemiceeuthanizedbycervicalvertebraedislocationunderanesthesiaonDay5.ThenscreenoutthemiceoftheexpressionofHoxa11up-regulatinganddown-regulatingsuccessfullythroughRealtimePCRandWesternblotonthelevelsofmRNAandprotein,atlastdetectedtheexpressionofIntegrin-β3，β-cateninandLif.Results:LevelsofIntegrin-β3,β-cateninandLifmRNAandproteinweredramaticallyincreasedanddecreasedinendometriumtransfectedsuccessfullywithpEGFP-Hoxa11andHoxa11SiRNAcomparedwiththecorrespondingcontrolones，consistentwiththeexpressionofHoxa11.Conclusion:Theexpressionofthefourfactorsmayinfluenceeachotherinmicro-environmentofendometrium.Hoxa11mayinfluenceendomtrialreceptivitythroughalteringtheexpressionofendometrialintegrin-β3，β-catenin,and
