EffectsofbaicaleinonTGF-β1-inducedfibroblasttomyfibroblastdifferentiation invitroAbstractEffectsof baicaleinonTGF-β1- inducedfibroblastto myfibroblast diferentiationinvitroAbstractObjective：Toinvestigatetheeffectsofbaicalein(BAE)ontransforminggrowthfactor-β1(TGF-β1）inducedfibroblastconverseintomyfibroblastanditsmechanism.Methods:HumanembryoniclungdiploidfibroblastWI-38wasinducedbyTGF-β1invitro,and thelevelsofalpha smooth muscleactin (α-SMA), CollagenI(ColI) weredeterminedbyWesternblotanalysis,cellmorphologywasobservedbyopticalmicroscopeandimmunohistochemistry.AfterbaicaleintreatmentofTGF-β1inducedWI-38fibroblastconverseintomyfibroblast,α-SMA、Smad2were detectedbyimmunohistochemistryandthechangesofmitochondria，golgimorphologywereobservedbytransmissionelectronmicroscope.Thelevelsofproteinα-SMA、ColI、Smad2、p-Smad2、Smad3、p-Smad3、Smad7weredeterminedbyWestornBlotanalysis,ThelevelsofmRNAα-SMA、Col-I、Smad2、p-Smad2、Smad3、p-Smad3、Smad7byReverseTranscriptionPCRmethod.Results:Comparedwithnormalgroup,2.5~40μg·L-1TGF-β1inducedfibroblastconverseintomyfibroblastaccordingtotimeanddosedependencytrendsinvitro.MTTshows:comparedwithnormalgroup,5μg·L-1TGF-β1inducedWI-38fibroblastsfor4days,Microscopeshow,cellproliferationwassignificantly,fromspindlefibroblastsorstarstructureintoaflatmusclefibrestructurefibroblasts,morphologicalchangesanddifferentiationwereobvious.accordingtoimmunohistochemistry,α-SMAwasonlyexpressedwithinnucleusinnormalgroup, andα-SMAwasexpressedboththenucleusandcytoplasmafter 5μg·L-1 TGF-β1 inducedfibroblastfor4days,Smad2wasinplentyofexpressioninthe nucleus.accordingtoWesternBlot,thelevels ofα-SMA、ColI wereup-regulatedfor4days,andtheresultwasinlinewithimmunohistochemistryresults.onlytoTGF-β1caninducedfibroblastconverseintomyfibroblast,andtheeffectcanlastfor3days.BAEintervention,Opticalmicroscopeobservations:TGF-β1 inducedWI-38celldifferentiation and cellproliferation was significantly inhibited; Immunohistochemical :Abstract Effectsofbaical
