Domestic and foreign research have found thatCOX-2overexpressedinmanytumorissuesascoloncarcinoma,andwasclosedrelevant withtumorigenesisanddevelopmentofcarcinomer.Celecoxibisofthesecondgeneration ofCycloxygenase-2inhibitor.Wetrytostudytheinfluenceofcelecoxibonthegrowth of HepG2invitro.Methods:(1)HepG2wasculturedintheenvironmentof37oC,5%CO2.Atthelogarithmic growthphase, thecell linewastreated by5 kindsofconcentrationgradient celecoxib as6.25，12.5，25，50，100μmol／l.(2)Making the culturesolutionofno drugsascontrol, andHepG2 celllinetreated bythesame concentrationofsolventsasblank,measuretheOD value at570nmthroughthemethodsofMTTafter24,48and 72hours’treatment.Calculate theinhibitionratesandmappingaccordingto theformula.(3)Makingthenormalcell line HepG2 ascontrol, evaluatethe apoptosisrateafter24htreatmentby5 differentkindsof concentrationofcelecoxib throughflowcytometryAnnexinV/PI. Analysistheresultsby software ModFitLT.(4)Making thenormalcelllineHepG2 ascontrol,assessthe cellcycle after24htreatmentby5 differentkindsofconcentrationofcelecoxibthroughflowcytometryPI.
