Arpincontributes tobacterialtranslocation anddevelopmentofsevere acutepancreatitisAbstractAIM:Thisstudyassessedthe impactofArpinproteinandtightjunction(TJ)proteinsintheintestinalmucosaonbacterialtranslocationinpatientswithsevereacutepancreatitis(SAP).METHODS:Fifty SAPpatientswereidentifiedasstudyobjectsand thenclassifiedintotwogroupsaccordingtothepresenceofbacterialtranslocation(BT)intheblood[i.e.,BT(+)andBT(-)].Twentyhealthyindividualswereincludedinthecontrolgroup.BTwasanalyzedbypolymerasechainreaction,colonicmucosaltissuewasobtainedbyendoscopyandtheexpressionofTJproteinsandArpinproteinwasdeterminedusingImmunohistochemistryandWesternblotting.RESULTS:BacterialDNAwasdetectedintheperipheralbloodof62.0%ofpatients(31/50)withSAP.TheexpressionofTJproteinsinSAPpatientswas lowerthanthatinhealthycontrols. Incontrast,ArpinproteinexpressioninSAPpatientswashigherthaninhealthycontrols(0.38±0.19vs0.28±0.16,P0.05).AmongSAPpatients,thosepositiveforBTshowedahigherlevelofclaudin-2expression(0.64±0.27vs0.32±0.21,P0.05)andalowerlevelofoccludin(OC)(0.61±0.28vs0.73±0.32,P0.05)andzonula occludens-1(0.42±0.26vs0.58±0.17, P=0.038)expressionincomparisonwithBT(-)patients.Moreover,thelevelofArpinexpressioninBT(+)patientswashigherthaninBT(-)patients(0.61±0.28 vs0.31±0.24,P0.05).CONCLUSION:Arpinproteinaffectsthe expression oftightjunctionproteinsandmayhaveanimpactonBT.Theseresultscontributetoabetterunderstandingofthefactorsinvolvedinbacterialtranslocationduringacutepancreatitis.Postgraduatestudent:WenshengDeng(GeneralSurgery)Directedbyprof.DianliangZhangKeywords:Severeacute pancreatitis; Arpin;Tightjunctionproteins;Bacterialtranslocation;Epithelium;Intestinalepithelialbarrier目录引言··············································································································································1第1章资料与方法··················································································································31.1研究对象·················································
