Experimentalstudyonmalignanttransformationofmacrophages inducedbygliomastemcellsAbstrcatPartI:Experimentalstudy onmalignanttransformationofhost macrophagesinducedbygliomastemcellsinvitroObjective:Hostmacrophageswereco-culturedwithgliomastemcells.Interactions betweenhumangliomastemcellsandhostmacrophageswereobserved,especially, whethermalignanttransformationofmacrophagesinducedbygliomastemcellscould occurwasfurtherstudied,anditsbiologicalphenotypiesaftertransformationwere identified.Method:C57BL/6J-NC-GFPnudemiceexpressinggreenfluorescentprotein(GFP) wereappliedtoharvestthemacrophageswithperitoneallavage.Gliomastemcell lines(SU3)wastransfectedwithredfluorescentprotein(RFP).SU3-RFPcellswere co-culturedwithmousemacrophagestoestablishadual-colorco-cultureinvitromodel. Theinteractionsbetweenthehost macrophagesandgliomastemcellswereobserved. Morphologicalchangesofthemacrophagecellswereobserved.Identificationofthe biologicalphenotypes oftransformedmacrophages wereperformed.Results:Macrophagesexperiencedmorphologicalchangesafterinvitrostimulation bytumorstemcells. phagocytosis oftumorcellsbymacrophageswas observedintheearly stageofcellco-culture.Thenmacrophagesturnedfromsmallroundcellsintolong spindle-likecells.Cellgrowthcurveshowedthatcellcycleandthegrowthrateofthe transformedmacrophagesweresimilartothoseofSU3.Thetransformedcellshadpatial cancercellcharacteristics.Conclusion：Dual-colorfluorescenceinvitrocellco-culturemodel canbeappliedtoobserveinteractionsbetweenhumangliomastemcellsandhostmacrophages.Malignant transformationofhostmacrophagescouldbeinducedbygliomastemcellsinvitrowhich deservedfurtherstudy.KeyWords:Thedual-fluorescencetracing;Malignanttransformation;Macrophages; GliomastemcellsPartIIPreliminarystudiesonmalignanttransformationofmacrophages inducedbygliomastemcellsinvivo.Objective:Recentresearchesontumorimmunityhaveshownthattumorinfiltrating macrophages playedpromotingrolesfavoredfortumorgrowthwhentumordevelopedinto theadvancedstage,thispaperaimstofurtherexplo
