Over time, if the suspended solid begins to separate in the centrifuge, more light passes through the sample. If the SC stable, the intensity of the light does not change. One method of testing the stability of an SC formulation is to apply high shear such as in a centrifuge. Shown is in the photo on the left is a Lumifuge (Lumi = light and fuge comes from centrifuge). The operation is relatively simple: the SC or emulsions or suspensions are placed in a sample chamber. The sample chamber is placed into the centrifuge and spun at 1 to 3000 rpm. There is a light source which is directed at the sample and transmission of the light through 20 millimeters height is determined by a detector on the opposite side of the sample chamber from the light.. Initially the specimen is very opaque and there is very low transmission. At periodic time intervals, the transmission of light through the sample is recorded. If the SC is stable, the transmission of light remains blocked and unchanged. If the SC is unstable, the solids begin to collected at the bottom of the sample chamber.
