APOBEC3A 核酸及表达蛋白水平基于生物信 息学的简略分析 万科星 2016120059 儿院 医学检验诊断学 wwwalies@163.com 摘要 APBEC3A(载脂蛋白 B mRNA 调控酶，催化样多肽 3A)属于 APOBEC3 家族，是多种正常 组织中突变的来源.近期研究发现。APOBEC3A 在多种肿瘤组织的基因组当中也具有强烈的 突变倾向。APOBEC3 家族是一类胞嘧啶脱氨酶，是固有免疫反应的一部分，具有抵抗细胞 内逆转录因子、逆转录病毒及 DNA 病毒的活性，而 APOBEC3A 具有与家族成员不同的一些 性质：抗病毒活性不同，可激活 DNA 损伤应答，导致细胞周期终止等。APOBEC 脱氨酶家 族可使管家基因脱氨基化，同时也可以引起核酸序列中 C→T 碱基突变，从而成为正常组 织中广泛的突变来源，并在病毒感染或过表达时诱导肿瘤组织的产生，从而使得 APOBEC3 家族成为潜在的治疗靶点。本文使用 NCBI 数据库下载获得 APOBEC3A 的核酸及蛋白序 列，对其启动子作一初步分析，并进行了同源建模，以期为后续的实验提供理论基础。 关键词：APOBEC3A，启动子分析，同源建模 Abstract APOBEC3A (apolipoprotein B mRNA editing enzyme, catalytic polypeptide-like 3A), a member of cytidine deaminase APOBEC3 family,which is a source of mutations in many normal is sues.Recent study showed that APOBEC3A also had a strongly tend of mutation in the genomes of many human cancers. APOBEC3 family is a family of cytidine deaminase,normally,this family members function as part of the innate immune system that protects against retrovirus and retrotransposon propagation.AlthoughAPOBEC3A performed some different features:this member of APOBEC3 family showed respective anti-virus activity,and could induce responds of DNA impairment ,leads to the end of cell cycle and soon.The APOBEC3 family also deaminate cytosines in the host genome and generate C→Tmutations,which could induce many human cancers when injected virus or over expression, brings APOBEC3A an posibility of cancer therapeutic target.To indentify the homology of APOBEC3A of APOBEC3 family and build a pre preparation of subsequent research, we download the nucleotide and protein sequence from NCBI,analysised the promoter of APOBEC3A and made homology modeling .
