AbstractTraditional Abstract Traditional Chinese medicine haS played irreplaceable role in modem healthcare system．Recently， modem medicine，chemistry and biology researchers focusing active ingredients ofChinese traditional medicine，as well mechanisms of these active ingredients in the treatment of diseases．Proteins the specific features oflife activity implementer，most drug molecules change the character ofspecifiC functional protein by interacting with the specific functional protein and thus play its due role． In thiS article,we selected the most numerous protein of the circulatory system，the human serum albumin(HSA)，as well pepsin，one ofthe main digestive en刁mes in the digestive system，as research subjects．Furthermore，we also select cyclovirobuxine D，helicid and aurantio。obtusfil from the three categories of the active ingredients which extracted from traditional Chinese medicine respectively，as another research subjects．We variety of methods，including steady-state fluorescence，synchronous fluorescence(SFS)，time．resolved fluorescence lifetime，Ultraviolet·Visble(UV‘vis)，Fourier transform infrared spectroscopy(FT-双)，circular dichroism(CD)，Molecular Modeling，to study and describe the t3q)e， character and some important parameters of the interaction between cyclovirobuxine D，helicid，aurantio。 obtusin mad HSA pepsin，besides，we also studied the difference and similarities ofthe interaction between aurantio-obtusin and HSA pepsin． Results and Conclusion：Fluorescence quenching data demonstrate that all of cyclovirobuxine D，helicid and aurantio．obtusin Can quench the fluorescence ofliSA pepsin，which means that all ofcyclovirobuxine D，helicid and aurantio．obtusin have interacted with HSA pepsin．S3mchronous fluorescence data indicats that cyclovirobuxine D，helicid and aurantio-obtusill quenched也e fluorescence ofprotein by interacting with the tryptophan residues and tyrosine residues，which include trp2 1 4 of HSA and trp39 of pepsin．The temperature control experiments show
