氨溴索增强左氧氟沙星在铜绿假单胞菌生物被膜形成时杀菌作用 【摘要】目的 观察盐酸氨溴索(AMB)对铜绿假单胞菌(P.a)的生物被摸(BF)破坏清除作用及其与左氧氟沙星(LFX)联合后对BF内P.a的影响。方法 分别建立PAO1的3 d和7 d体外BF模型,随机(随机数字法)分为空白对照组、AMB组,AMB+LFX组。AMB质量浓度分别为256 μg/ml和512 μg/ml。于体外早期和成熟BF模型形成后,分别加入不同质量浓度的AMB,作用24 h后,连续稀释法行载体表面BF内P.a活菌计数,SEM观察载体表面BF形态学变化; 采用单因素方差分析法(One-Way ANOVA)进行计量资料的统计分析。结果 无论是早期BF还是成熟期BF,药物作用24 h后SEM观察,AMB组载体表面BF较空白对照组明显减少,并且随着AMB质量浓度的增加,载体表面BF呈减少趋势。无论是早期BF还是成熟期BF,1 μg/ml的 LFX组可以减少BF内细菌(P 【关键词】铜绿假单胞菌;生物被膜;盐酸氨溴索;左氧氟沙星;杀菌作用 Ambroxol reinforce the bactericidal effect of levofloxacin on the Pseudomonas aeruginosa biofilm KONG Jin-liang, CAI Shuang-qi, CHEN Yi-qiang, YAN Ping, ZHANG Dong-wei, JIAN Li-juan,WU Hai-ying. To evaluate the synergistically bactericidal effect of AMB along with levofloxacin (LFX)on BF of P.a.Methods The early model (cultured for 3 d) and mature model (cultured for 7 d) of P.a wild strain (PAO1) BF were established, in vitro, respectively. The models were randomly(random number)divided into control group, AMB group and AMB+LFX group. The concentrations of AMB were 256 μg/ml and 512 μg/ml, respectively. When the early BF model and mature BF model were made, different concentrations of AMB were added in AMB group and AMB+LFX (1 μg/ml) was added in AMB+LFX group. The number of viable P.a on the BF carrier was counted with the continuous dilution method 24 h after AMB or/and LFX added.
