AbstractTwonovelBODIPYfluorescentdyesweredesignedandsynthesized,onthebasisofpreviousexperiments.AnovelpHprobeandaratiometricfluorescentprobeforClO?withcoumarinfluorophoreweredesignedandsynthesizedbasedonICTmechanism.Thispaperisdividedintofourchapters,includingthefollowing:Thefirstchapterisaliteraturereviewsection,whichdemonstratesthegenerationoffluorescence,thesensingmechanismofsmall-moleculefluorescentprobes,sometypicalfluorescentpHprobesandhypochloriteprobes.ThesecondchapterisconcerningthedesignandsynthesisofnewBODIPYdyesandtheirapplicationsforbio-imaginginlivingcells.Themeso-COOEtsubstitutedBODIPYswasdesignedandsynthesized,whichexhibitedhighquantumyieldandredtofar-redemission.Notably,themeso-COOEtsubstitutedBODIPYsdisplayedalmostseparatedUV-VisabsorptionandemissionspectrawithalargeStokesshift(~60nm).Time-dependentdensityfunctionaltheorycalculationswereconductedtounderstandthestructure–opticalpropertiesrelationship,anditrevealedthatthelargeStokesshiftwasresultedfromthegeometricchangefromthegroundstatetothefirstexcitedsingletstate.Furthermore,BODIPYwastestedforitsabilitytoimageinlivingcells.TheresultsindicatedthatDye-1wasawater-solubleandmembrane-permeableprobe.ThethirdchapterisconcerningthedesignandsynthesisofnewpHfluorescentprobeanditsapplicationforcellimaging.WehavedevelopedahighlysensitiveandselectivepHprobethatrespondstobasicpHwithcolorimetricandfluorescenceturn-onsignal.Interestingly,theprobeNaOCydisplaysextremelylargeStokesshift(~122nm),whichcaneffectivelyavoidauto-fluorescencefromcellularspecies.AccordingtoUV-visspectraandfluorescencetitration,agoodlinearitywasobtainedandthepKais8.32.Inaddition,goodphotostability,finereversibilityandwatersolubilityin100%PBSsolutionmakeitfavorableforintracellularpHimaging.TheconfocalfluorescentmicroscopicimagesofsubcellulardistributionindicatesthatNaOCyhasgoodcellmembranepermeabilityandcouldselectivelystainnucleus,whichisthefirstexampleofpHprobewithsuchfeature.Thefourthchapterisconcerningthedesignandsynthesisofnewfluorescent
